[0078] a. Mixed Species Biofilm Test :mix species Bacteria were grown as follows: Actinomyces viscosus (ATCC# 31346). for mixing species Determination, the source of the bacteria was inoculated with Actinomyces viscosus, Lactobacillus casei (Lactobacillus casei) (ATCC#334), Streptococcus oralis (ATCC #35037), Fusobacterium nucleatum (ATCC#10953 ) and Veilonella parvula (ATCC #17745) continuous culture chemostat. This mixed culture was maintained in a dedicated complex medium at 37°C in a continuous culture chemostat.

[0079] Mixing performed on saliva-coated HAP discs (scHAP) species biofilm. After saliva coating, discs were inoculated with 1 ml of the mix from the above continuous culture chemostat species The culture, which was diluted with 0.5x TSB to an OD610 of ~0.2. Plates were incubated at 37°C for 48 hours to allow biofilm formation.

[0080] Discs containing pre-formed biofilms were transferred to new 24-well culture plates and 1 ml of sterile 0.5x TSB ...

Abstract

Described herein are oral care compositions comprising (i) an orally acceptable gallium salt and (ii) a basic amino acid polymer, in amounts effective to disrupt biofilm; and methods of making and using the same.

Description

Background of the invention [0001] While many antimicrobials are tested against free-floating, planktonic cultures of bacteria, these organisms rarely exist in this way in nature. More commonly, bacteria are found in complex communities called biofilms. This is especially the case in the oral cavity, where up to 700 species of bacteria form complex and mixed species biofilms, commonly known as dental plaque. Plaque can lead to a variety of undesirable conditions including erosion of tooth enamel, tooth hypersensitivity, bad breath, tooth decay, dental caries and gingivitis. Bacteria found in biofilms differ in their metabolic rates, nutrient access, and requirements and sensitivities to antimicrobial agents relative to free-living bacteria. Agents active against bacteria in culture may be relatively ineffective against bacteria in plaques without some mechanism for disrupting biofilms. [0002] Gallium is a transition metal usually found in the form of gallium(III) salts. ...
