Problems solved by technology

[0006] Since strawberry is mainly asexually propagated through stem-growing seedlings in production and application, the excellent characteristics of new strawberry varieties are seriously affected, and the cycle of stem-moving seedling breeding is long, which restricts the demonstration application and variety promotion of "Jiuxiang" strawberry

Embodiment 1

[0025] Example 1 Explant disinfection and shoot tip growth point stripping

[0026] 1. See figure 1 , Select a strong and disease-free "Jiuxiang" strawberry mother plant with a terminal bud of about 1.5 cm in size on the stolon, dip it in a small amount of washing liquid and gently scrub it with your hands for 2 to 3 times, rinse it with running water for 90 to 120 minutes, and then put it on the ultra-clean workbench. Rinse with 75% alcohol for 10 seconds, wash with sterile water for 3 times, sterilize with 0.5% sodium hypochlorite for 7 minutes, rinse with sterile water for 6 to 7 times, and finally blot the surface moisture with sterile filter paper.

[0027] 2. Peel off the leaves of the outer layer of the terminal bud layer by layer under sterile conditions and under a dissecting microscope, and cut off the growth point with a leaf primordia (about 0.2-0.3 mm in length) with a scalpel (see figure 2 ), immediately inoculated on the differentiation medium (recipe: MS+6-BA...

Embodiment 2

[0028] Rooting, acclimatization and transplanting of the multiplication culture of embodiment 2 clustering buds and tissue culture seedlings

[0029] 1. After four weeks, clustered buds grow from the tip of the stem. When the buds grow to 2 cm in size, cut off the buds one by one and divide them into individual plants. Put 5 to 6 buds in each bottle, and then transfer them to the proliferation medium (formula: MS+6 -BA0.2mg / L+IBA0.02mg / L+white sugar 30g / L+agar 8g / L, pH=5.7~5.8) to continue to proliferate, culture temperature 24±1℃, 12 hours / day light nourish. The multiplication coefficient reaches 3, and the statistical results are shown in Table 1 and Table 2 below. One generation is multiplied every 28 days, and a large number of non-toxic seedlings are obtained after multiplication of 6 to 7 generations. See the multiplication results for image 3 .

[0030]2. Cut the strawberry sprouts cultured through subculture from the bud cluster, and transfer them to the rooting med...

Abstract

A "Jiuxiang" strawberry stem tip rapid breeding method by tissue culture and virus removal is disclosed. The rapid breeding method includes: explant disinfection, stem tip tissue virus removal and culture, multiplication culture, rooting culture, seedling hardening and transplanting. A stem tip tissue culture method is adopted to breed "Jiuxiang" strawberry virus-free seedlings, thus ensuring that all the excellent characteristics of parents are stably inherited, and therefore influences of outside conditions can be prevented, and the rapid breeding method can be performed at all seasons, thus saving the seedling culture land, and reducing the production cost. A large quantity of good test-tube plantlets can be produced in a short period of time. Large-scale production can be performed to provide "Jiuxiang" strawberry seedlings with a large quantity and good quality for the market so as to meet market needs. The "Jiuxiang" strawberry virus-free seedlings prepared by the rapid breeding method are high in growth vigor, disease-resistant and increased in big-fruit yield. The yield can be increased by 30-50%.

Description

technical field [0001] The invention belongs to the field of plant tissue culture, and relates to a method for rapid propagation of "Jiuxiang" strawberry stem tip tissue culture detoxification. Background technique [0002] In recent years, with the increase of strawberry continuous cropping years and the promotion of susceptible varieties such as "Fengxiang" and "Hongyan", the incidence of strawberry anthracnose has shown a sharp increase in China. Generally, after the susceptible seedlings are planted in the field, about 80% of the plants will die as they grow by next March. In southern regions such as Shanghai, the strawberry fruit is harvested from December of the current year to May of the next year, which lasts up to six months, with a large number of harvests in February, March, and early April. In this way, strawberry growers will suffer huge losses. Therefore, it has become an urgent problem to promote new disease-resistant strawberry varieties comparable to straw...
