Abstract

The invention provides a culture medium for inducing cyclamen leaf to form callus and bud and a culture method. Formation of cyclamen callus and bud is highly-efficiently induced by a two-step method, and explant browning is effectively inhibited. The culture method comprises the following steps: before acquisition of explant leaves, cyclamen plant undergoes shading treatment for 24-26 h, and afterwards, in vitro low-temperature (20-22 DEG C) darkness culture is carried out so as to effectively solve the browning problem; then, callus is induced through low-temperature darkness culture, culture temperature is raised to 25-28 DEG C after 30-40 days, and light culture is carried out to induce bud formation; and finally, root induction is conducted after bud has been formed for 15-20 days. The technical system can effectively inhibit browning, has high propagation coefficient, can preserve original traits and heterosis of F1 generation, and provides key technical support for factory production of cyclamen seedlings.
