The aqueous bi-phase system comprises the following raw materials by mass: 50 to 70.1 percent of n-butylpyridium chloride ionic liquid, 15.3 to 24.2 percent of monopotassium phosphate and the balance of water, wherein the total percentage of the three ingredients is 100 percent. The extraction rate of vitamin B6 from watermelon pulp can reach up to 98.9 percent according to the application. The application has the advantages that the linearity range is wide; the detection limit is low; the relative standard deviation is relatively low; the detection recovery rate to a sample is high; the extraction effect is favorable; a large quantity of vitamin B6 can be provided for various fields such as biological medicine due to the aqueous bi-phase system and the application.
