Embodiment 1

[0050] Mix 3.00g of 1-ethyl-3-methylimidazolium phosphate dimethyl salt ionic liquid, 3.13g of n-butanol and appropriate amount of distilled water to make the total mass of the system 10g; add 2g of the red star apple peel grinding liquid to be tested , and then ultrasonically oscillated for 30 min under the conditions of temperature 35° C. and pH 5.5, phase separation and static extraction for 24 h.

[0051] In order to test the effect of the present invention on separating and extracting the flavonoids in the red star apple peel, the content of the flavonoids was measured, and the extraction rate was 97.2%.

Embodiment 2

[0053] The preparation method in this example is the same as that in Example 1, the only difference being that the conditions in the preparation process are different. In this example, the ionic liquid is changed to 1-ethyl-3-methylimidazolium tetrafluoroborate, and the temperature changes 65°C, and the rest of the conditions remain unchanged.

[0054] In order to test the effect of the present invention on separating and extracting the flavonoids in the red star apple peel, the content of the flavonoids was measured, and the extraction rate was 80.7%.

Embodiment 3

[0056] The preparation method in this example is the same as that in Example 1, the only difference being that the conditions in the preparation process are different. In this example, n-butanol is changed to an equivalent amount of isobutanol, the pH value becomes 3.0, and the rest of the conditions remain unchanged. .

[0057] In order to test the effect of the present invention on separating and extracting the flavonoids in the red star apple peel, the content of the flavonoids was measured, and the extraction rate was 50.2%.

Abstract

The invention discloses an aqueous two-phase system and applications thereof in flavone separation from 'Starking' apple peel. The aqueous two-phase system comprises a 1-ethyl-3-methylimidazolium dimethyl phosphate ionic liquid, n-butanol and water. The aqueous two-phase system comprises 24.5-35.0% by mass of the 1-ethyl-3-methylimidazolium dimethyl phosphate ionic liquid and 27.5-43.0% by mass of the n-butanol, with the balance being the water and the sum of mass percentages of the ionic liquid, the n-butanol and the water being 100%. The aqueous two-phase system is applied for flavone separation from the 'Starking' apple peel and the extraction rate of flavones reaches 97.2%. The aqueous two-phase system s is characterized by a wide linear range, a low detection limit, low relative standard deviation and a high recovery rate of samples, and is simple in operation, high in extraction rate and suitable for quantification research on the flavones in the 'Starking' apple peel.

Description

technical field [0001] The invention belongs to the field of food science and engineering, and in particular relates to a two-phase aqueous system and its application for separating flavonoids from red star apple peels. Background technique [0002] In recent years, due to the development of free radical life sciences, total flavonoids, which have strong antioxidant and free radical elimination effects, have received unprecedented attention. Total flavonoids are involved in the metabolism of phosphoric acid and arachidonic acid, phosphorylation of protein, transfer of calcium ions, scavenging of free radicals, enhancement of antioxidant activity, redox, chelation and gene expression. Bioflavonoids from natural sources have a small molecular weight, can pass through the blood-brain barrier, enter adipose tissue, and be quickly absorbed by the body. Their health benefits are: (1) anti-inflammatory (2) anti-allergic (3) inhibits bacteria (4) inhibits parasites (5) inhibits vir...
