Embodiment 1

[0029] A method for measuring the toxicity of pesticide fungicides by model organism algae of the present invention comprises the following steps:

[0030] S1. Preparation of test liquid:

[0031] Preparation of BG11 medium dilution: Prepare the BG11 medium formula according to the formula in Table 1. The preparation method is: prepare the ingredients in Table 1 to make the corresponding mother solution concentration, and transfer the corresponding mother solution dosage to the 1000mL volumetric flask according to the indicated order and distilled water to 1000mL, autoclaved at 121°C for 15 minutes, sealed and labeled, stored in a refrigerator at 4°C, and valid for 2 months. Dilute the BG11 medium 10 times with distilled water to obtain the BG11 medium dilution.

[0032] Weigh 0.1000 g of 50% difenoconazole suspension, dissolve it with the BG11 medium dilution and dilute it to a 100 mL volumetric flask to obtain a stock solution with a concentration of 500 mg a.i. / L. Pipette...

Embodiment 2

[0054] A method for measuring the toxicity of pesticide fungicides by model organism algae of the present invention comprises the following steps:

[0055] 1. Test liquid preparation:

[0056] Pipette 0.625mL of 80 g / L tebuconazole suspension seed coating agent, dissolve and settle to 100mL volumetric flask with BG11 culture medium dilution (the composition of BG11 dilution is identical with embodiment 1), obtain concentration and be 500mg a.i. / L stock solution. Pipette 0.300, 0.600, 1.200, 2.400, 4.800mL of the above stock solution in sequence, respectively dilute to 250mL volumetric flask with BG11 culture medium dilution, and prepare the concentration of 0.600, 1.20, 2.40, 4.80, 9.60mg a.i. / L 2 times the test liquid.

[0057] S2, the preparation of algae dilution: measure the cell concentration of algae stoste (Cercis cerevisiae) with hemocytometer method and be 4.00 * 10 6 , and diluted 100 times with sterile water to obtain the algae dilution, the concentration of alga...

Abstract

The invention discloses a mode biological algae method for determining pesticide bactericide toxicity. The method comprises the following steps: preparing a pesticide bactericide into a test liquid with gradient concentration; preparing a bio-algae solution into an algae diluted liquid with sterile water; mixing the test liquid with the algae diluted liquid, culturing for 72 h under sealing condition; counting the algae cell number at every 24 h, and recording the biological effect parameters of the algae cells and the physicochemical parameters of the test liquid; and according to the biological effect parameters calculating the median effective concentration EC50 of the pesticide on the algae cells and determining the toxicity of the pesticide bactericide. The method has the advantages of easy operation, strong practicability, short period and easiness to judge end point.

Description

technical field [0001] The invention relates to the field of cultivation technology, in particular to a method for measuring the toxicity of pesticide fungicides by model algae. Background technique [0002] As the primary producers of aquatic ecosystems, algae play an extremely important role in the balance and stability of water bodies. Algae testing in biological testing is an essential method in aquatic ecotoxicology research. After pesticides are used in the field, they will migrate into the water body and affect the growth of algae, thus affecting the entire ecosystem. Algae are often used in the research of algae growth inhibition toxicity test in the registration of new chemical substances and in the research of algae growth inhibition toxicity test in pesticide registration. [0003] At present, there are few research reports on the toxicity screening of pesticides and fungicides at home and abroad. Even if there are some reports and related research models, there ...
