Abstract

An analysis and identification method for distinguishing different sea cucumber varieties is disclosed. The method includes a step of performing spicule identification by a microscope, namely a step including (1) immobilizing collected sea cucumbers with absolute ethanol, (2) cutting body wall tissues having a size of 1 cmZ X 1mm from the middles of backs of the sea cucumbers, with the tissues being adopted as analysis samples, (3) preparing three repeated analysis samples for each sample, cleaning the samples with double-distilled water, absorbing water with sterile water absorbing paper until the samples are dry, and adding the samples to small beakers having a volume of 5 mL (with each sample being provided with an separate beaker and a separate pipet so as to prevent mutual contamination, and 4) adding dropwise 1 mL of a NaClO solution having a concentration of 100o and digesting for 30 s; and a step of sample preparing for a scanning electron microscope. Identification is performed by a method of microscope spicule differences assistant by the scanning electron microscope. Under the microscope, spicules having different shapes can be found, the spicules are an important character gradually completed and stabilized along with sea cucumber embryonic development, shapes and sizes of the spicules are greatly different for different varieties, and the spicules are an important basis for sea cucumber classification and rapid identification.
