Problems solved by technology

Breeders have always wanted to introduce the heat-resistant genes in Yuanjiang wild rice into cultivated rice through interspecific hybridization, but the breeding of heat-resistant varieties has been limited due to linkage cumbersomeness and the difficulty of stabilizing hybrid offspring.

Embodiment 1

[0022] Embodiment 1 selects parents

[0023] Jiangxi Provincial Academy of Agricultural Sciences Rice National Engineering Laboratory (Nanchang) Super Rice Research and Development Center used Shuhui 527 as the recurrent parent and Yuanjiang Wild Rice Hehuatang 3 as the donor parent (non-recurrent parent), and self-crossed in the 5th generation of backcross Combined with the SSR molecular marker-assisted selection covering the whole genome in the third generation, a set of 112 single-segment replacement lines of Yuanjiang common wild rice with the background of Shuhui 527 were bred, and the genome coverage rate reached 73.5%. In 2010, on the first day after rice heading, plants with consistent growth were selected and put into pots and moved into artificial climate chambers. Three plants were planted in each pot, and three replicates were set up for each material, and the water and fertilizer were managed conventionally. Set the relative humidity to be 85%, the light to be 12h / ...

Embodiment 2

[0025] (1) In the summer of 2011, a row of YJ07‐04‐06 and 9311 were planted at the Nanchang Experimental Station of Jiangxi Academy of Agricultural Sciences, 9311 (CGMCC NO.12532) was the female parent, and YJ07‐04‐06 (CGMCC NO.12534) was the father F 1 , all mixed harvest after maturity. Planting 3 rows of F in Sanya, Hainan in the winter of 2011 1 , backcross to obtain BC 1 f 1 , all mixed harvest after maturity. Field management was carried out by conventional methods.

[0026] (2) Molecular marker-assisted selection: In the summer of 2012, 50 rows of BC were planted at the Nanchang Experimental Station of Jiangxi Academy of Agricultural Sciences 2 f 1 , 8 plants in each row, and the collected leaves of each plant were put into 2ml eppendorf tubes, filled with small steel balls, and then added 600 μl of 1.25% SDS extract solution after high-throughput tissue grinder high-speed vibration grinding, and the SDS method was used to extract DNA. The molecular markers (Tabl...

Abstract

The invention belongs to the field of molecular seed breeding and relates to a molecular seed breeding method for breeding a new paddy rice variety which has heat resistance during heading and flowering stage. The method includes the steps of: 1) with a mid-indica two-line elite restorer line 9311, which is assigned the accession number of CGMCC No.12532, as a female parent, and a wild rice chromosome fragment introgression line YJ07-04-06, which is assigned the accession number of CGMCC No.12534, as a male parent, performing hybridization for 1 generation and backcross for 3 generations, and then performing selfing for 3 generations to obtain BC3F4, wherein each generation is harvested in a single plant manner; 2) identifying and selecting a new paddy rice variety by means of molecule marker assisted selection with combination of phytotron for simulating high temperature stress. By means of the method, a new paddy rice variety "Yuanye No.2" having heat resistance during the heading and flowering stage has been cultured. Target characters are selected through molecule marker assistance, so that a restorer line having heat resistance during the heading and flowering stage can be cultured within 3-4 years quickly and high-effectively.

Description

technical field [0001] The invention belongs to the field of molecular breeding, and relates to a molecular breeding method for improving the heat resistance of rice heading and flowering stage by using a single-segment replacement line, in particular to a molecular breeding method for improving the heat resistance of rice heading and flowering stage by using a single-segment replacement line of wild rice method. Background technique [0002] High temperature heat damage is one of the main factors affecting the yield and quality of rice in the world, and it is also the main natural disaster of rice cultivation in China. It mainly occurs in the south of the Yangtze River Basin (Xiong Zhenmin, editor-in-chief Cai Hongfa. China Rice. Beijing: China Agricultural Science and Technology Press, 1992 ). . High temperature disorder at the heading stage of rice (also known as heat damage) refers to the phenomenon that rice spikelet pollination and fertilization cannot be carried out...
