Embodiment 1

[0068] Embodiment 1: Analysis of the skin whitening effect of the peptide of the present invention

[0069] Cells were treated with various concentrations of the peptide of SEQ ID NO: 1 or SEQ ID NO: 2, and then the activity in melanogenesis inhibition was measured. 5% CO at 37°C 2 Mouse melanocytes were cultured in DMEM supplemented with 10% FBS in an atmosphere. Cells were seeded in 24-well plates (1X10 5 cells / well) and confirm attachment. Cells were incubated for 3 days with each test group: (a) solvent only, as a negative control; (b) 20 μg / ml α-MSH, as a positive control; (c) 20 μg / ml α-MSH plus 1, 10, 100 and 150 μg / ml peptide. After centrifugation, the medium was removed and the level of melanogenesis was observed visually.

[0070] The results showed that the peptide had an inhibitory effect on melanogenesis in a concentration-dependent manner. Compared with the α-MSH treatment group, the color of the medium in the peptide treatment group of the present inventio...

Embodiment 2

[0072] Example 2: Synergy

[0073] Contains 1X10 5 Mouse epidermal keratinocytes and 5X10 4 A 2:1 mixture of darkly pigmented mouse epidermal melanocytes was seeded per well in a 24-well cell culture plate to establish a co-culture system. After overnight incubation, the cells were treated with fresh medium containing the peptide of the invention and incubated for 72 hours after treatment. The medium was then removed and replaced with 80 [mu]l dimethylsulfoxide (DMSO). Plates were further incubated with DMSO at 65° C. for 1 hour, after which the well contents were ground and the same samples were used for melanin content assay (read at 450 nm in a plate reader) and protein evaluation (BiCinchoninic Acid method). The percentage reduction in melanin content is relative to the untreated control. The results are shown in Table 1 below.

Embodiment 3

[0077] Embodiment 3: the preparation of whitening cream

Abstract

The invention relates to an active peptide with a skin whitening effect and a whitening cream containing the active peptide. Particularly, the invention discloses the active peptide containing peptides of SEQ ID NO:1 and SEQ ID NO:2 and the whitening cream containing the active peptide, and the two peptides are independently used or used in a combined manner for skin whitening. The whitening cream comprises the following components by weight percent: 2.0-4.0% of glutathione, 0.5-2.5% of vitamin E, 0.5-2.5% of rose hip seed oil, 2.0-4.0% of a glycyrrhiza glabra extracting solution, 4.0-6.0% of a mulberry leaf extract, 2.0-4.0% of 1,3-propanediol, 4.0-6.0% of sodium hyaluronate, 1.5-3.5% of rose essential oil, 2.0-4.0% of soybean lecithin, 5.0-15.0% of resveratrol, 0.4-0.8% of carbomer, 2.0-4.0% of glucan and 2.0-4.0% of a tremella extract. The whitening cream has the effects of tightening skin, whitening and the like.

Description

technical field [0001] The present invention relates to non-natural peptides having skin lightening effects and their use in cosmetic and personal care compositions. [0002] Background of the invention [0003] As of now, various natural and synthetic peptides have been found to be widely used in cosmetic compositions. Generally, peptides are included in cosmetics for their functional activities, such as enzyme inhibition, antiviral and antibacterial activities. [0004] US Patent 6,620,419 discloses cosmetic compositions comprising polypeptides, such as Lys-Thr-Thr-Lys-Ser, which are reported to induce collagen and glycosaminoglycan synthesis for the treatment of skin aging. [0005] US Patent 6,821,524 describes the use of the small naturally occurring polypeptide thymosin-beta-4 (TB4) in cosmetic compositions to improve the appearance of aged or damaged skin. Vascular endothelial growth factor (VEGF) and transforming growth factor beta 1 (TGFβ) peptides are also said to...
