[0030] Example 1 Construction of an Improved Model of Painful Diabetic Peripheral Neuropathy in Rats

[0031] 1. Diabetic rat model

[0032]Adult male Sprague-Dawley rats (purchased from the Experimental Animal Center of Xinhua Hospital Affiliated to Shanghai Jiao Tong University School of Medicine, and the Experimental Animal Center of Shanghai Academy of Biological Sciences), with an initial weight of 200-230 g, were purchased from the Experimental Animal Center of Xinhua Hospital Affiliated to Shanghai Jiao Tong University School of Medicine. Raising was carried out under the feeding conditions of a laminar flow animal house, with a feeding temperature of 22-24°C, a regular 12-hour day and night cycle, 5 rats / cage, and sufficient water and food.

Abstract

The present invention relates to a method for constructing an improved rat model of painful diabetic peripheral neuropathy. The construction method is as follows: (1) establishing a diabetic rat model by intraperitoneally injecting streptozotocin at a dose of 60 mg/kg once; 2) An annular ferrule is arranged on the sciatic nerve of the diabetic rat, and the annular ferrule surrounds the sciatic nerve, contacts it but does not oppress it. The present invention creatively adopts the sciatic nerve entrapment method, puts the latex tube on the sciatic nerve of diabetic rats, and constructs a rat model of painful diabetic peripheral neuropathy. The experimental results show that the probability of MA occurring in rats in the STZ injection + latex tube placement group Compared with STZ-injected rats, it was significantly increased (P<0.05). The method of the invention has the advantages of short modeling time, high modeling rate and stable model, and provides a basis for the research on the mechanism of diabetic peripheral neuropathy pain and related medicines.

Description

technical field [0001] The invention relates to the technical field of animal model preparation, in particular to a method for constructing an improved rat model of painful diabetic peripheral neuropathy. Background technique [0002] Diabetes mellitus (DM) is a clinically common chronic metabolic disease, and diabetic peripheral neuropathy (DPN) is a common chronic neurological complication of DM, with a prevalence rate as high as 47% to 91%. The incidence of diabetic peripheral neuropathy (PDPN) can reach 13% to 26%. It is a common phenotype of DPN, which brings great hardship to patients, and even leads to serious consequences of amputation and death in diabetic patients. Common clinical manifestations of PDPN include symmetrical numbness, pain, burning sensation, chills, hypoesthesia, aggravated in the afternoon or night, and neuropathic pain characterized by allodynia, spontaneous pain, and hyperalgesia . PDPN patients often suffer from depression, sleep disorders, an...
