Embodiment Construction

[0018] The present invention generally relates to compositions and methods for the treatment of lysogenic viruses and lytic viruses. These compositions can treat lysogenic viruses and lytic viruses, or optionally viruses using both replication methods.

[0019] As used herein, "lysogenic virus" refers to a virus that replicates through the lysogenic cycle (ie, does not cause host cell rupture and integrates viral nucleic acid into host cell DNA). Lysogenic viruses can replicate mainly through the lysogenic cycle, but sometimes through the lytic cycle.

[0020] As used herein, "lytic virus" refers to a virus that replicates through a lytic cycle (ie, causes the host cell to rupture after virus accumulation in the cell). The lytic virus can replicate mainly through the lytic cycle, but sometimes through the lysogenic cycle.

[0021] "GRNA" as used herein is guide RNA. The gRNA in the CRISPR Cas9 system herein is used to excise viral genome segments, and therefore severely destroy th...

Abstract

A composition for treating a lysogenic virus, including isolated nucleic acid encoding two or more gene editors chosen from gene editors that target viral DNA, gene editors that target viral RNA, andcombinations thereof. A composition for treating a lytic virus, including isolated nucleic acid encoding at least one gene editor that targets viral DNA and a viral RNA targeting composition. A composition for treating both lysogenic and lytic viruses, including isolated nucleic acid encoding two or more gene editors that target viral RNA, chosen from CRISPR-associated nucleases, Argonaute endonuclease gDNAs, C2c2, RNase P RNA, and combinations thereof. A composition for treating lytic viruses, including isolated nucleic acid encoding two or more gene editors that target viral RNA and a viralRNA targeting composition. Methods of treating a lysogenic virus or a lytic virus, by administering the above compositions to an individual having a virus and inactivating the virus.

Description

Technical field [0001] The present invention relates to compositions and treatment methods for viruses. More specifically, the present invention relates to compositions and therapies for excising and inactivating viruses from infected host cells. Background technique [0002] The virus replicates through one of two cycles: the lytic cycle or the lysogenic cycle. In the lysis cycle, the virus first penetrates the host cell and releases its own nucleic acid. Next, the metabolic mechanism of the host cell is used to replicate the viral nucleic acid and accumulate the virus in the host cell. Once enough virus particles are produced in the host cell, the host cell ruptures (lysis) and the virus particles continue to infect other cells. A lytic virus can integrate viral DNA into the host genome and is non-integrating, where lysis does not occur during cell infection. [0003] Splitting viruses include JC virus (John Cunningham virus) (JCV), hepatitis A, and various herpes viruses. I...
