Embodiment Construction

[0031] The present invention will be described in further detail below in conjunction with the accompanying drawings and embodiments.

[0032] The present invention uses real-time fluorescent quantitative PCR to detect the copy number variation of the goat MYLK4 gene and is used for molecular breeding, comprising the following steps:

[0033] (1) Use the NCBI database to find the MYLK4 gene sequence, then use the Primer5 software to design primers, and use ordinary PCR to detect the primers;

[0034] (2) Use real-time fluorescent quantitative PCR (qPCR) technology to detect the copy number variation of candidate sites in the population, and screen out CNV markers related to goat growth traits;

[0035] (3) Use SPSS 23.0 software to conduct association analysis between the type of copy number variation and the growth traits of goats;

[0036] (4) Breeding goats with excellent growth traits according to the type of copy number variation.

[0037] 1. Goat sample collection

Abstract

The invention discloses a detection method and application of goat MYLK4 gene CNV marker. The method is based on real-time fluorescent quantitative PCR, using goat genomic DNA as a template to amplify the copy number variation region of the goat MYLK4 gene, and then using ‑ΔΔCt to calculate and determine the type of copy number variation of the individual based on the results. The present invention utilizes individual CNV types and growth traits for association analysis. The detection method provided by the invention lays a theoretical foundation for the establishment of the relationship between the copy number variation of the MYLK4 gene and the growth traits of goats. The method is simple, fast, easy to popularize and use, and is conducive to accelerating the establishment of goat populations with excellent germplasm resources.

Description

technical field [0001] The invention belongs to the field of genetic breeding, in particular to a method for detecting MYLK4 gene copy number variation (CNV) markers and its application in molecular breeding for improving growth traits of goats. Background technique [0002] With the rapid development of related disciplines such as genomics and bioinformatics, the theory and technology of animal genetics and breeding have also undergone major changes, that is, the traditional conventional phenotypic breeding has gradually shifted to molecular breeding. At present, research on molecular breeding is mainly focused on marker-assisted selection (MAS), which is a technology that selects genetic resources or breeding materials through DNA molecular markers, and improves the comprehensive traits of livestock and poultry. In livestock and poultry breeding, through the selection of DNA markers that are closely related to growth traits and quantitative traits, the purpose of early sel...
