Abstract

The invention discloses a primer for a CDS (Coding Sequence) domain sequence of an ACP5 (Acid Phosphatase 5) gene of clonal chicken. The invention designs a group of specific primers, PCR (PolymeraseChain Reaction) amplification is carried out on the ACP5 gene of chicken by combining the primers and applying an RT (Reverse Transcription)-PCR technology, a target fragment of the PCR amplificationis a complete CDS domain sequence of the ACP5 gene of the chicken, the obtained complete CDS domain sequence of the ACP5 gene is high in accuracy and reliable in result, and basic materials are provided for constructing protokaryotic and eukaryotic expression vectors of the ACP5 gene of the chicken, related animal models and the like. According to the primer disclosed by the invention, high-GC (Guanine-Cytosine) content sequence cloning is obtained through a common cloning means; enzyme cutting sites are added to the primer, so that the primer can be directly used for other expression experiences, and meanwhile, an important laboratory significance in treating and improving bone metabolism of caged laying hens is obtained.
