According to the method, a silkworm cocoon of a specific bombyx mori silk fibroin missing type mutant variety is adopted, a sericin water solution is extracted, ultrasonic treatment is performed, and the sericin hydrogel with new characteristics is obtained. The sericin hydrogel has hightransparency; the sericin hydrogel has a microstructure different from conventional hydrogel, hydrogel micropores are orderly arranged from large to small and from small to large; the functional sericin hydrogel has the characteristics of excellent elasticity, injectability and controllable gelling time; the sericin hydrogel has certain application value in the aspect of drug release control; thesericin hydrogel has higher affinity for cells and can support adhesion and proliferation of the cells; the sericin hydrogel and a 3D porous sericin biological scaffold obtained through freeze dryingof the sericin hydrogel can serve as extracellular matrixes to support cell growth and promote exchange of nutritional substances and can be applied to repairing of damage of multiple tissues and treatment of diseases.
