Problems solved by technology

However, due to the complex composition of dodder water extract, the dodder water extract contains anthraquinone, coumarin, flavonoids, glycosides, sterols, tannic acid, sugars, etc., which is difficult to be internationally recognized. The use of compounds to improve sperm motility and enhance human sperm acrosome reaction is a technical problem to be solved in this field

Embodiment 1

[0033] Example 1 F10-Evaluation of the effect of the composition containing asthenoside on the improvement of sperm motility in severe asthenozoospermia

[0034] 1. Sample Preparation

[0035] Sperm samples from patients with severe asthenozoospermia were selected for the experiment. Semen ejaculation by masturbation, using a specific sterile container to collect sperm samples after 2-7 days of abstinence. The sample water bath was kept at 22-33°C, and the semen after liquefaction was evaluated according to the WHO1999 evaluation standard, using the computer-assisted semen analyzer CASA, to evaluate the sperm parameters (volume, sperm count, percentage of motility and motility characteristics). Asthenozoospermia is defined as grade A sperm count <25% or A+B <50%, (World Health Organization, 1999), while all other parameters are normal. Specimens containing white blood cells or somatic cell contamination were excluded based on morphological observations. The motility paramet...

[0045] Example 2 F10-Evaluation of the effect of the composition containing asthenosperm on improving the motility of sperm in moderate to mild asthenozoospermia

[0046] 1. Sample Preparation

[0047] Sperm samples from patients with moderate to mild asthenozoospermia were selected for the experiment. Semen ejaculation by masturbation, using a specific sterile container to collect sperm samples after 2-7 days of abstinence. Embodiment 3

[0056] Example 3 F10-Evaluation of the effect of the composition containing ascarin on the sperm motility of normal sperm

[0057] 1. Sample Preparation

[0058] Normal sperm samples donated by healthy men were selected for the experiment. Abstract

The invention relates to a composition containing astragalin and used for improving the fertilization ability and acrosome reaction of human sperms. The composition comprises a buffer solution and astragalin. The human sperms purified after separation are cultured, in addition to maintaining sperm motility, the fertilization ability and acrosome reaction of normal human sperms can be significantlyimproved, and a meaningful reference is provided for improving assisted reproductive technology. The astragalin is added in the buffer solution, so the fertilization ability of sperms in patients with clinical asthenospermia can be significantly improved, the improvement degree of the fertilization ability is most obvious in severe asthenospermia, the fertilization ability of some cultured spermsreaches the normal range of the fertilization ability of normal sperms, and the improvement degree in severe asthenospermia is most obvious. Meanwhile, the acrosome reaction of the sperms is significantly increased. The result fully demonstrates that the use of astragalin can improve the low fertility of sperms in patients with asthenospermia.

Description

technical field [0001] The invention relates to a composition containing ascarin, in particular to a composition for improving the fertilization ability of in vitro cultured sperm and its application. Background technique [0002] A survey on infertility organized by the World Health Organization in the mid-to-late 1980s showed that the infertility rate in developed countries was 5% to 8%, and in some places in developing countries it even reached 30%. According to incomplete statistics, infertility patients account for about 2% to 6% of Chinese couples of childbearing age, and there is a gradual increase trend. The emergence of human assisted reproductive technology has gradually solved the problem of infertility that has plagued the medical field for many years and brought joy to countless families. Human assisted reproductive technology refers to the technology of using eggs, sperm, fertilized eggs and embryos to cure the symptoms of infertility. At present, it mainly i...
