The method comprises the following steps: S1, dissolving a plutonium sample to prepare a plutonium isotope diluent solution whose plutonium concentration and plutonium isotope components are predetermined; S2, adsorbing a plutonium dissolving solution to be detected by using resin to obtain resin for adsorbing the plutonium dissolving solution to be detected; S3, measuring the plutonium isotope ratio of the resin for adsorbing the plutonium dissolving solution to be detected obtained in the S2 by utilizing a gamma spectrum; calculating and determining plutonium isotope components in theplutonium dissolving solution to be detected according to the measured plutonium isotope ratio; S4, mixing the plutonium isotope diluent solution with a plutonium dissolving solution to be detected to obtain a mixed solution, and adsorbing the mixed solution by using resin to obtain resin for adsorbing the mixed solution; S5, determining the plutonium isotope ratio of the resin for adsorbing themixed solution obtained in the S4 by utilizing gamma spectroscopy; and S6, carrying out calculating to determine the plutonium concentration in the plutonium dissolving solution to be detected.
