Problems solved by technology

[0006] However, the chemical and physical properties of Puer tea fermented in piles and urban sewage plant sludge and farmland soil are highly heterogeneous, and the number and population distribution of microorganisms are also significantly different. difference
At present, there are no relevant reports on the microbial community composition of compost-fermented Pu-erh tea using PCR-DGGE technology at home and abroad

Abstract

The invention relates to an identification method and a special primer of a dominant fungal population in a pile-fermentation process of puer tea, wherein the identification method comprises the following steps of: (1) acquiring puer tea samples to be detected in different fermentation processes; (2) extracting the total DNA (Deoxyribose Nucleic Acid) of the samples to be detected; (3) carrying out PCR (Polymerase Chain Reaction) amplification under the guide of the special primer by taking the total DNA of the samples to be detected as a template; (4) carrying out denaturing gradient gel electrophoresis (DGGE) on a target segment, and dyeing by use of sybrgreen to obtain a DGGE fingerprint spectrum; and (5) recovering a dominant strip, carrying out PCR amplification under the guide of the special primer by taking the dominant strip as a template, and sequencing the target segment; and (6) carrying out sequence comparison and analysis on a sequencing result to determine the structure of the dominant fungal population in the pile-fermentation process of the puer tea.

Description

technical field [0001] The invention relates to an identification method of dominant fungal flora in the stacking fermentation process of Pu'er tea and special primers thereof. Background technique [0002] Pu-erh cooked tea is a post-fermentation made from the sun-dried green tea of Yunnan's unique big-leaf tea [Camellia sinensis (Linn) var. tea. Microbes play an important role in the process of fermenting Pu-erh tea. Without microorganisms, the quality and flavor of Pu-erh tea cannot be guaranteed. This invention studies the dominant microbial flora in Pu-erh tea fermented by stacking and An identification method is provided. If these dominant flora are used in the stacking fermentation production process of Pu'er tea, it will be beneficial to the production of Pu'er tea and improve the quality. [0003] At present, the research on microorganisms in fermented Pu-erh tea mainly uses traditional methods such as separation, purification and identification of microorganism...
