Abstract
The invention relates to an identification method and a special primer of a dominant fungal population in a pile-fermentation process of puer tea, wherein the identification method comprises the following steps of: (1) acquiring puer tea samples to be detected in different fermentation processes; (2) extracting the total DNA (Deoxyribose Nucleic Acid) of the samples to be detected; (3) carrying out PCR (Polymerase Chain Reaction) amplification under the guide of the special primer by taking the total DNA of the samples to be detected as a template; (4) carrying out denaturing gradient gel electrophoresis (DGGE) on a target segment, and dyeing by use of sybrgreen to obtain a DGGE fingerprint spectrum; and (5) recovering a dominant strip, carrying out PCR amplification under the guide of the special primer by taking the dominant strip as a template, and sequencing the target segment; and (6) carrying out sequence comparison and analysis on a sequencing result to determine the structure of the dominant fungal population in the pile-fermentation process of the puer tea.
