Embodiment 1

[0030] Embodiment 1: A kind of measuring method of inferring the moth moth time of silkworm

[0031] (1) Materials and equipment

[0032] The silkworm chrysalis was a variety bred by Suzhou University (Suxiu×Chunfeng); the incubator was a 250D artificial climate box produced by Guangdong Shaoguan Xinteng Science and Technology Instrument Co., Ltd.; the quantitative PCR instrument was ABI ViiA7.

[0033] Samples: More than 6 male pupae or female pupae were collected every 48 hours for the construction of the model.

[0034] (2) Stacking and quantity collection

[0035] The silkworms were reared in the upper cocoon by the conventional method, the cocoons were picked after 3 days in the cocoon, the cocoons were cut open and the unpupated silkworms were taken out. After 2 hours after the silkworms pupated, the blood of 6 male silkworm chrysalis or female silkworm chrysalis was collected respectively. Three replicates were set up, and this pupal developmental stage was recorded a...

Abstract

The invention discloses a method for judging the moth formation time of bombyx mori. The method comprises the following steps of: (1) independently constructing an associated model of pupal period development time and a relative transcription expression quantity of the (Cocoonase,Coc) gene of a male pupa and a female pupa of a variety to be detected; and (2) measuring the relative expression quantity of the (Cocoonase,Coc) gene of the pupa to be detected, and according to the gender, contrasting with the associated model obtained in (1) to carry out calculation to obtain the pupal period development time so as to judge the moth formation time. By use of the method, the pupal period development time of one or a plurality of pupae is accurately measured so as to accurately judge the moth formation time of the bombyx mori, an error caused by traditional visual inspection can be reduced, and a new detection method is provided for judging the moth formation time in bombyx mori species manufacturing work.

Description

technical field [0001] The invention relates to a method for judging the moth moth time of silkworm, in particular to an accurate detection method for pupal stage development time of silkworm based on the relative transcription expression level of genes. Background technique [0002] silkworm( Bombyx mori ) is an economically important insect that has been domesticated in China for more than 5,700 years. Silkworms go through four different developmental stages throughout their life, namely egg, larva, pupa and adult. The metamorphosis and development process from the pupal stage to the adult stage is called eclosion, and this process generally takes 10 to 15 days, and the cocoons are usually picked about 7 days after being put on the cocoon. The silkworm does not need to take in nutrients from the external environment during the pupal stage, but the destruction of larval tissues and organs is violently carried out in the body, and the formation of adult tissues and organs ...
