The method comprises the steps of S1 separating lymphocytes from ex-vivo blood provided by a subject, culturing the lymphocytes in vitro for stimulation and cell staining, detecting specific surface protein on the surface of a single lymphocyte in the lymphocytes, and detecting and calculating the number and proportion of various cells according to positive or negative results displayed by different specific surface proteins; S2 performing standardized scoring on the number and the proportion of each type of cells to obtain a corresponding standard score; S3 classifying the functions represented by the cell types corresponding to the scores into six types, namely T cell reserve, an inherent immune system, an NK cell killing function, T cell balance, T cell activation and T cell functions; summarizing the scores of the index items contained in each type to obtain a total score of the type; and S4 summing the six types of total scores of a testee to obtain an analysis result corresponding to the blood sample of the certain testee.
