[0065] In some embodiments, the device is configured for disposable, portable, single use, inexpensive molecular diagnostic methods. The device may include one or more modules configured to perform high quality molecular diagnostic tests including, but not limited to, sample preparation, nucleic acid amplification (eg, via polymerase chain reaction, isothermal amplification, etc.), and detection. In some embodiments, sample preparation can be performed by isolating pathogens / entities and removing unwanted amplification (eg, PCR) inhibitors. The target entity can then be cleaved to release the target nucleic acid for amplification. A target nucleic acid (eg, a target polynucleotide sequence) in a target entity can be temperature cycled with a polymerase or amplified via isothermal incubation to generate a larger number of copies of the target nucleic acid sequence for detection.
