Benefits of technology

The technical effect of this patented solution involves reducing harm caused by storing it under specific conditions while combining its ingredients together for better use. This helps prevent damaging changes from happening overtime or affecting other components' performance.

Problems solved by technology

Technological Problem addressed in this patents relates to improving long term storability (LTS) during donor'bloods due to decreased activity or deterioration caused by factors like temperature fluctuate overtime. Current methods involve storing red blood plasma with other substances such as albumin instead of reducing its effectiveness. However, these techniques also cause harmful side effects when stored under certain conditions.

Embodiment 1-4

[0016] See Table 1 below for the components and contents of the red blood cell preservation solution, and the test results about time and hemolysis rate in Examples 1-4 are as follows figure 1 shown.

[0017]

[0018] Table 1

[0019] From figure 1 It can be seen that Example 1, which added sodium bicarbonate and haptoglobin at the same time, has the longest storage time and the best storage effect, and after 100 days, the hemolysis rate basically no longer increases; Example 3 with only haptoglobin added Next; Only add the embodiment 2 of sodium bicarbonate again; The hemolysis rate of the embodiment 4 that sodium bicarbonate, haptoglobin all do not add is always higher than embodiment 1-3.

Embodiment 5-8

[0021] See Table 2 below for the components and contents of the red blood cell preservation solution, and the test results about time and hemolysis rate in Examples 5-8 are as follows figure 2 shown.

[0024] Embodiment 5-8 about the test result of time and hemolysis rate as figure 2 Shown, conclusion is with embodiment 1-4.

Embodiment 9-12

[0026] See Table 3 below for the components and contents of the red blood cell preservation solution, and the test results about time and hemolysis rate in Examples 9-12 are as follows image 3 shown.

[0029] Embodiment 9-12 about the test result of time and hemolysis rate as image 3 Shown, conclusion is with embodiment 1-4.

[0030] After analysis, the present invention adds haptoglobin Hp, which can specifically combine with free hemoglobin FHb in blood, so that NO in red blood cells will not be oxidized to nitrate at a high speed, and the ability of cells to transport oxygen is improved, thereby improving red blood cells. Survival time; at the same time, adding carbonate to the buffer system can effectively reduce low-temperature storage damage (HSLs), which not only improves the shelf life of reagent red blood cells, but also improves the quality of red blood cells during the test, making the test results more accurate. close to the real r...

Abstract

The invention provides a red blood cell preserving fluid, which adopts a phosphate and carbonate mixed buffer system and comprises the following parts in content: 30 to 40 mg/mL of water glucose, 0.3 to 0.5 mg/mL of adenine, 8 to 12 mg/mL of mannitol, 0.03 to 0.05 mg/mL of disodium hydrogen phosphate, 0.05 to 0.15 mg/mL of sodium bicarbonate, 5 to 15 mg/mL of sodium citrate, 0.3 to 0.7 mg/mL of chloramphenicol and 2 to 4 mg/mL of sodium chloride. According to the invention, carbonate is added into a buffer system of the red blood cell preserving fluid, so that low-temperature storage damage can be effectively reduced.
