The method takes crude fluorescein as a raw material, and is characterized in that pure water pulping and recrystallization are carried out after acetylation reaction, and then sodium methoxide is used for deacetylation refining to obtain refined fluorescein. The invention relates to a novel process for purifying fluorescein by utilizing a method for preparing a fluorescein derivative. Fluorescein diacetate is easy to prepare and reduce, has very good crystallinity and is greatly different from the impurities in property, so that the problems of repeated use of acid and alkali and coprecipitation of the impurities and fluorescein in the prior art are solved by a purification route of preparing fluorescein diacetate from fluorescein sodium and then carrying out saponifying and reducing.
