Benefits of technology

This patented technology allows researchers to quickly identify signs or symptoms associated with diseases like hepatitis B virus-related illnesses by analyzing blood samples from their own body without causing any harmful side effect on healthy individuals nearby. It's highly sensitive but requires expensive equipment such as an assay plate made up of specialized materials designed specifically against certain genes found at high levels in these indicators.

Problems solved by technology

This patented technical problem addressed in this patents relates to improving understanding how Hepatocyte growth factors (HEMCF)/microRNAs (miRNets) control different aspects during liver diseases such as virilization and fibrogenesis processes involved in livers'diseas including cholangiolar sialadencer transport proteins (CSLT-1 ). These interactions between components contribute to their functions like stabilizing membranous structure and controlling mitochondrial activity. Therefore, identification of specific markers associated with inflammatory cytokines and other targets could lead to earlier discovery of highly sensitive and non invasively detected targeted therapies for treatment purposes.

Embodiment 1

[0063] Example 1 Cloning and identification of methylated ELF gene promoter regulatory region

[0064] The tumor suppressor gene ELF is expressed in normal liver tissue, while its expression is down-regulated in liver cancer stem cells and liver cancer tissues, and knocking out the ELF gene in mice leads to spontaneous liver cancer, indicating that ELF plays an important role in the early stage of liver cancer. The regulatory mechanism of ELF gene silencing under natural conditions is still unclear. The present invention explores the relationship between the methylation state of the CpG island in the promoter region of the ELF gene promoter region and HCC from the perspective of DNA methylation, and lays an experimental foundation for further analysis of the methylation state of the ELF gene promoter region in tumor tissues and body fluid samples .

[0065] In this example, the regulatory sequence of the ELF gene promoter region (ELFpro, 435 bp) was amplified by PCR method, a...

Embodiment 2

[0089] Example 2 Study on the relationship between the methylation status of the CpG island in the promoter region of the ELF gene and liver cancer

[0090] 1 Materials and methods

[0091] 11 cell lines

[0092] What used in this experiment is all conventional cell lines that have been reported in the art. The purpose of using these cell lines in this experiment is to serve as positive control cell lines for CpG island methylation, to illustrate the present invention, and to implement the technical solution of the present invention. independent of these cell lines. Instead of using these cell lines, other cell lines that have been reported that the CpG island of the corresponding gene has been methylated can also serve the above role, and even the sequence of the CpG island of the corresponding gene that has been reported can be used. Refer to the method in Example 1 , to prepare a positive template for the complete methylation of the gene promoter region, or to prepare a p...

Abstract

The technical problem to be solved in the invention is providing a reagent, wherein the reagent is used to detect a methylation of a CpG island which is a cancer suppressor gene starting region in an off-body sample. The technical scheme solving the technical problem in the invention is providing a kit for early warning and screening reagent for liver cancer. The kit comprises a reagent, by which the methylation state of a CpG island that is a gene starting region in a spectrin of a human embryonic can be detected. By using the kid provided in the invention, if the methylation of ELF gene is independently detected, the detection rate can reach 64.70%. And the invention further shares the kit with other detecting reagents for detection of the methylation of the CpG island that is a cancer suppressor gene starting region, and a novel composite detection kit is obtained, of which the detection rate can reach 93.55%. The kit in the invention and a kit of serum AFP400 can be used together in practice to obtain a more accurate effect.
