Embodiment Construction

[0026] The seed embryo paraffin sections of the mixed collection of Paeonia lactiflora seeds were successfully prepared.

[0027] combined with Figure 1-3 , the specific implementation is as follows:

[0028] 1. Materials: The peony mixed seeds were harvested in late August 2012.

[0029] 2. Pretreatment of seeds

[0030] (1) Wash the seeds: wash with detergent for 10-20 minutes, and rinse with running water.

[0031] (2) Screening of seeds: water selection, after soaking the seeds for 2-3 hours, remove the floating unfilled seeds.

[0032] (3) Sterilize the peony seeds with 0.5% potassium permanganate solution for 40 minutes, and rinse them with distilled water.

[0033] (4) Soak the seeds in warm water at 45°C for 48 hours in an incubator to soften the seed coat and allow the seeds to absorb water.

[0034] (5) Remove the floating unfilled seeds.

[0035] (6) 15 ℃ sand storage, take the peony seeds of 0 d, 30 d and 60 d in the sand, wash them, use absorbent paper to a...

Abstract

The invention discloses a production method for a paraffin section of a paeonia lactiflora mature embryo. The production method comprises the following steps: drawing materials and fixing: stripping a seed coat, cutting a seed, immersing the seed top part containing the embryo or part endosperm into a FAA stationary liquid, and fixing for more than 24 hours; dehydrating: washing in ethyl alcohol with the concentration of 70% for 3 to 5 times, and performing gradient dehydration from the ethyl alcohol with the concentration of 70%; performing hyalinization: carrying out five level hyalinization by using dimethyl benzene as a transparent reagent; waxing and embedding: pouring out one half volume of dimethyl benzene, adding wax crumbs in a bottle until saturation, staying overnight at the temperature of 40 DEG C; then changing a wax liquid every two hours for three times per day, repeating for three days, and carrying out conventional paraffin embedding; slicing; flattening and bonding: flattening a linoleum tape on an iron plate of a water bath kettle at the constant temperature of 50 DEG C, dewaxing, rehydrating, dyeing through safranine and fast green and performing hyalinization; mounting. According to the invention, the paeonia lactiflora mature embryo paraffin section of which the tissue structure is integrated and clear can be obtained, so that accurate technical support can be provided for researching paeonia lactiflora seed dormancy and germination.

Description

technical field [0001] The invention relates to a method for preparing paraffin sections, in particular to a method for preparing paraffin sections of mature seed embryos of peony. Background technique [0002] peony ( Paeonia lactiflora ) is a perennial perennial flower of the genus Paeonia in the family Paeonia, and it is a traditional famous flower in China with a long history of cultivation. [0003] During the long-term systematic evolution, the seeds of Paeonia suffruticosa developed special dormant germination characteristics, namely the dormancy characteristics of epicotyl and hypocotyl. Seed propagation is the only way to obtain peony seedlings and is the basis of asexual reproduction. Under natural conditions, it takes 6-7 months from sowing to emergence, and the growth cycle is long, which brings great difficulties to the artificial cultivation and production of peony and the inspection of seeds. Exploring this special dual dormancy mechanism is of great signif...
