Embodiment 1

[0022] 1. Sample pretreatment (DNA extraction)

[0023] 1. Take about 0.01g of the back muscle of 5 hairtail samples, numbered dy1, dy2, dy3, dy4, dy5, put them into a 1.5ml Buffer tube, cut them up with sterile small scissors, add 400μL of SDS extraction buffer and 8μL of 20mg / mL Proteinase K, mixed evenly, placed in a water bath at 55°C for 2 hours or overnight at 37°C, shaking several times in between.

[0024] 2. Add 300 μL of 6M / L NaCl solution to the supernatant, shake well, and centrifuge at 10,000 g / min for 30 min.

[0025] 3. Transfer the supernatant to a new tube, add an equal volume of chloroform:isoamyl alcohol (24:1) mixture, shake well, and centrifuge at 10000g / min for 10min.

[0026] 4. Transfer the supernatant to a new tube, add an equal volume of isopropanol, precipitate at -20°C for more than 1 hour, and centrifuge at 10,000 g / min for 10 minutes.

[0027] 5. Discard the supernatant, wash the precipitate twice with 70% alcohol and once with absolute ethanol....

Embodiment 2

[0036] 1. Sample pretreatment (DNA extraction)

[0037] 1. Take about 0.01g of the back muscles of 5 large yellow croaker samples, numbered dhy1, dhy2, dhy3, dhy4, dhy5, put them into a 1.5ml Buffer tube, cut them into pieces with sterile small scissors, add 400μL of SDS extraction buffer and 8μL of 20mg / mL Proteinase K, mixed evenly, placed in a water bath at 55°C for 2 hours or overnight at 37°C, shaking several times in between.

[0038] 2. Add 300 μL of 6M / L NaCl solution to the supernatant, shake well, and centrifuge at 10,000 g / min for 30 min.

[0039] 3. Transfer the supernatant to a new tube, add an equal volume of chloroform:isoamyl alcohol (24:1) mixture, shake well, and centrifuge at 10000g / min for 10min.

[0040] 4. Transfer the supernatant to a new tube, add an equal volume of isopropanol, precipitate at -20°C for more than 1 hour, and centrifuge at 10,000 g / min for 10 minutes.

[0041] 5. Discard the supernatant, wash the precipitate twice with 70% alcohol and ...

Abstract

The invention provides a method for detecting marine products of the same origin. The method comprises the following steps: designing a specific primer by utilizing a CO1 sequence of cytochrome oxidase genes in mitochondrial genes of a group of marine products of the origin, performing PCR amplification on the different extracted marine products, and generating nucleotide sequences of different PCR fragments; comparing different gene sequences with the CO1 gene sequence of the marine products of the origin, performing biological analysis, thereby determining the authenticity of the products. A rapid, accurate and sensitive authentication method is established for protecting the geographical indication products. According to collection and amplification comparison of a group of conserved genes in aquatic products, molecular bar codes are marked on the geographical indication aquatic products, so that the origin is identified, the local fishery resources are protected, and the competitiveness and popularity of local fishery are improved.

Description

technical field [0001] The invention belongs to seafood detection and relates to a detection method of seafood from the same origin. technical background [0002] Geographical indication certification trademarks are an internationally accepted system that provides proof of origin and product quality and can increase the attractiveness of products to consumers. In other words, geographical indications mean that the quality is far from comparable to other similar products. The use of geographical indications to certify trademarks can not only protect local unique natural resources, products and cultural heritage, but also help locals implement brand fishery development strategies, improve the competitiveness of local fisheries, and greatly enhance local popularity. In real life, it is not uncommon for commercial aquatic products to be mislabeled, especially for those profitable aquatic products. Some unscrupulous traders often pass off local aquatic products as aquatic produ...
