Embodiment Construction

[0072] The present invention will be described in further detail below in conjunction with the embodiments and accompanying drawings.

[0073] see figure 2 As shown, the protein secondary mass spectrometry identification method based on the Poisson distribution model in this embodiment, the specific steps are as follows:

[0074] (1) Virtual enzymolysis protein database sequence, and establish peptide database and peptide database index for the peptide after enzymolysis according to the mass number of the peptide;

[0075] (2) According to the nucleus-to-mass ratio of the precursor ion in the experimental spectrum to be analyzed, find out the candidate peptides that meet the requirements in the peptide database described in step (1), and select the candidate peptides that meet the requirements as the theoretical spectrum;

[0076] (3) Carry out isotopic peak removal and denoising processing (that is, select effective peaks) for the experimental spectrum to be analyzed;

Abstract

The invention discloses a tandem mass spectrometry identification method for protein based on a Poisson distribution model. The method roughly comprises the following steps: fabricating an enzymolysis protein data bank sequence, establishing a peptide fragment data bank index for peptide segments having undergone enzymatic hydrolysis, and constructing a peptide fragment data bank; then finding out candidate peptide fragments meeting requirements from the constructed peptide fragment data bank according to the mass number of parent ions in to-be-analyzed experimental map after removal of charges; subjecting the experimental map to removal of isotopic peaks and de-noising processing; matching and marking the to-be-analyzed experimental map and a theoretical map in the data bank and selecting a peptide fragment with a highest mark as the identification result of the experimental map; and subjecting the identification result to integral false positive control. The tandem mass spectrometry identification method for protein based on the Poisson distribution model can dynamically choose peaks and has fast operation speed; and an effective map number and an effective peptide fragment number identified by the method are higher compared with algorithms in the prior art.

Description

technical field [0001] The invention relates to the field of protein secondary mass spectrometry identification, in particular to a protein secondary mass spectrometry identification method based on a Poisson distribution model. Background technique [0002] Biomass spectrometry has become one of the supporting technologies for proteome research. The development of mass spectrometry can be traced back to the parabolic mass spectrometer created by J.J.Thomson in the early 20th century. After that, the speed-focused mass spectrometer made by Aston in 1919 created a miracle for the development of mass spectrometry. Then, in the late 1980s, the emergence of two soft ionization techniques, matrix-assisted laser desorption ionization (MALDI) and electrospray (Electrospray Ionization, ESI), introduced fewer impurities into biological mass spectrometry while maintaining the purity of peptide molecules. Integrity, these changes make biological mass spectrometry technology can be app...
