Problems solved by technology

Phaeodactylum tricornutum is currently used industrially for the production of omega-3 polyunsaturated fatty acids, but this application, as well as others such as biofuels, is limited when TAG accumulation is induced by conventional nutrient deficient methods such as nitrogen starvation. Industrial implementation is still limited by growth retardation and low yields of biomass (Chisti, Journal of Biotechnology 167(2013) 201-214)
These methods have an important drawback: nitrogen deficiency leads to growth limitation

[0105] Phaeodactylum tricornutum (Pt1 ) Bohlin strain 8.6 CCMP2561 (Type Collection of Marine Organisms, now known as NCMA: National Center for Marine Algae and Microbiota) was used in each experiment.

[0106] Pt1 was grown at 20°C in a 250 mL flask in "enriched artificial seawater" (ESAW) medium prepared according to the regulations of the Canadian Center for the Culture of Microorganisms.

[0107] To prepare ESAW media, four separate solutions were prepared: two salt solutions (Solution 1 and Solution 2), one nutrient solution and one vitamin solution. Add salt to distilled deionized water (DDW). When the salts in Solution 1 and Solution 2 are completely dissolved, mix Solution 1 and Solution 2 together. Dilute the total volume with DDW.

Abstract

The present invention relates to a method of accumulating triacylglycerols in microalgae by inhibiting sterol metabolism by incubating the microalgae with inhibitors of sterol metabolism. The present invention also relates to a method for the production of fatty acids, biofuels, pharmaceutical or cosmetic compositions, and food supplements, said method comprising the step of accumulation of triacylglycerols in microalgae according to the present invention. Finally, the present invention focuses on the use of inhibitors of sterol metabolism to accumulate triglycerides in microorganisms, and preferably microalgae.

Description

technical field [0001] The invention relates to a method for accumulating triacylglycerol in microalgae by inhibiting sterol metabolism. The invention also relates to a method for the production of fatty acids, biofuels, pharmaceutical or cosmetic compositions, and food supplements, said method comprising the step of accumulating triacylglycerols in microalgae according to the invention. Finally, the present invention concerns the use of inhibitors of sterol metabolism for the accumulation of triglycerides in microorganisms, and preferably microalgae. Background technique [0002] It is known that crops used for nutritional purposes cannot be converted to crops used for oilseed production (Durrett et al., The Plant Journal (2008) 54, 593-607). Therefore, efforts have been made to perform oil production in other organisms such as algae (Chisti, Biotechnology Advances 25 (2007) 294-306; Chisti, Trends in Biotechnology, 2008, Vol 26, No. 3; Dismukes et al., Current Opinion in ...
