Abstract

The invention relates to a cryopreservation method of pollen of amur honeysuckle; method comprises: picking fresh pollen of amur honeysuckle from a well-developing parent tree at 13:30 on the right day of blossoming, loading to a 1.8 ml cryogenic tube, storing in a liquid nitrogen tank a -196 DEG C, the cryogenic tube holding the pollen when initial water content is about 13-14% after picking, and thawing for 5 min with tap water. The method of the invention is simple to perform, uses small space, is high in safety and stability and low in storage cost and provides greatly extended storage time for pollen so that storing pollen for long time is made possible; mature pollen contains the information of all genetic materials of a plant, and the research on cryopreservation of pollen is significant to the researches on accelerating the breeding process of amur honeysuckle and completing germplasm resource storage methods for amur honeysuckle, as well as other researches.

Description

technical field [0001] The invention relates to a process for long-term preservation of pollen, in particular to a process for ultra-low temperature preservation of honeysuckle pollen. Background technique [0002] Honeysuckle, also known as honeysuckle wood, chicken bone tree, etc., is a small deciduous tree in the Lonicera family Lonicera, often clustered into shrubs, with beautiful flowers and fruits, and has high ornamental value. The berries gradually change from yellow to red in June and July. The red turns purple. In gardens, honeysuckle bushes are often planted on lawns, hillsides, forest edges, roadsides, or dotted around buildings. They are good ornamental shrubs. Honeysuckle is a rare medicinal ornamental shrub, which has antipyretic, immune-enhancing and antibacterial effects. The whole plant can be used as medicine, which can dispel wind and detoxify, reduce swelling and relieve pain, and mainly treat dizziness and bruises. The stem bark can be used to make art...
