Abstract

The invention belongs to the field of macroalgae bioassay and particularly relates to a method for identifying four kinds of main enteromorpha green algae (enteromorpha, enteromorpha compressa, enteromorpha flexuosa and enteromorpha linza) by utilizing gel electrophoresis. The method provided by the invention comprises the following steps: (1) extracting total DNA of an algae body sample; (2) respectively performing PCR amplification through a 5S rDNA spacer sequence and an ITS sequence; (3) identifying by agarose gel electrophoresis, and analyzing and judging green tide enteromorpha algae species under strip conditions through observing an electrophoresis band and a position thereof after the end of the electrophoresis. The method provided by the invention has the benefits that the experimental operation is simple, and the green tide enteromorpha algae is not required to be sent to a sequencing company for sequencing; therefore, the cost is lower, the time consumed is short, and the data is accurate.
