Embodiment 1

[0059] Example 1 Inhibition of B.plantarii toxin secretion by thiadiazole compounds

[0060] 1 Culture of B. plantarii

[0061] The Burkholderia plant B. plantarii strain was isolated from the paddy fields of Zhuji, Zhejiang, and was cultured daily in a constant temperature incubator at 25°C. The medium used was nutrient broth agar medium.

[0062] 2 Inoculation of B. plantarii

[0063] Pick up a single colony of B.plantarii vertically with an inoculation needle, place it in 50ml nutrient broth medium for pre-culture, and after 24 hours of cultivation, draw 10ml of bacterial liquid, centrifuge at 5000rpm for 5min, discard the supernatant, and then add Rinse with an equal amount of clean water, shake and mix well, then centrifuge at 5000rpm for 5 minutes, add the same amount of clean water again and shake and mix well, absorb 100 μL of the mixed bacteria solution and add it to 60ml MA liquid medium for cultivation, the formula of MA medium Ratio for inorganic salt solution (N...

Embodiment 2

[0073] Embodiment 2 growth matrix addition method

[0074] 1 rice germination

[0075]Select a sufficient number of rice seeds, use 2% sodium hypochlorite to carry out surface disinfection for half an hour, and then place them in a sterilized glass petri dish after fully washing, and then add a small amount of sterilized water (subject to not submerging the surface of the seeds), Then put them in a constant temperature incubator at 25°C for cultivation, and replace the sterilized water every other day. After 2-3 days, the seeds will appear white, and the germination is completed at this time.

[0076] 2 Inoculation of B. plantarii

[0077] Pick up a single colony of B.plantarii vertically with an inoculation needle, place it in 50ml nutrient broth medium for pre-culture, and after 24 hours of cultivation, draw 10ml of bacterial liquid, centrifuge at 5000rpm for 5min, discard the supernatant, and then add Rinse with an equal amount of clean water, shake and mix, then centrifu...

Embodiment 3

[0097] Embodiment 3 seed treatment method

[0098] 2-amino-5-mercapto-1,3,4-thiadiazole solution (0.005mM-5mM), 2,5-dimercapto-1,3,4-thiadiazole solution (0.005mM-5mM), 2-mercapto-5-methyl-1,3,4-thiadiazole solution (0.005mM-5mM) and 0.01% dimethyl sulfoxide solution are used for germination acceleration, and the rice seeds after acceleration germination are divided into four batches. Each batch was added with the prepared B. plantarii suspension as the inoculation group or an equal volume of sterilized water as the non-inoculation group (subject to not submerging the seed surface), and cultivated for 24 hours. Planted in MS plant medium, 12 per bottle. The culture bottle was placed in a constant temperature incubator at 25°C, and the rice seedlings were cultivated under 12 hours of light and 12 hours of darkness every day, and their growth conditions were continuously observed, and the lengths of roots and leaves of rice seedlings were measured on the third day for statistic...

Abstract

The invention discloses application of a thiadiazole compound in the preparation of a medicament for preventing and treating bacterial seedling damping-off of rice. A preparation prepared by using the thiadiazole compound as a main effective component is capable of effectively inhibiting the level of tropolone produced by B. plantarii, and therefore, the bacterial seedling damping-off of the rice can be prevented and treated. The medicament provided by the invention can be utilized for simply and feasibly preventing and treating the bacterial seedling damping-off of rice, is low in cost and has good prevention and treatment effects.

Description

technical field [0001] The invention relates to the field of pesticide science, in particular to the application of thiadiazole compounds in the preparation of medicaments for preventing and treating bacterial blight of rice seedlings. Background technique [0002] Burkholderia plantarii is one of the important pathogens causing bacterial blight of rice seedlings. It has strong infectivity, fecundity and adaptability, and seriously threatens rice production. In nature, B. plantarii is often distributed in seedbed soil, field water, plant seeds, and animal and plant residues. After entering the seedling field, it completes the initial infection under suitable conditions, and enters the re-infection cycle through seed and soil transmission. Infected seedlings showed dwarfing, chlorosis and root growth disorder in the early stage, and withered and died in the later stage. [0003] Pathological studies have found that B. plantarii secretes a small molecular virulence factor cal...
