Embodiment 1

[0039] Weigh 0.35g chitosan and dissolve in 35mL dilute acid solution to obtain chitosan aqueous solution. Dissolve arginine and leucine (50:1 molar ratio) in 15 mL of buffer solution and activate for 10 hours to obtain an amino acid activation solution with an amino acid concentration of 300 mg / mL. The amino acid activation solution was mixed with chitosan aqueous solution and reacted for 24 hours. After the reaction is finished, the product is purified to obtain the chitosan derivative with selective antibacterial properties.

[0040] According to the method requirements of GB / T 16886.5-2003 "Biological Evaluation of Medical Devices Part Five In Vitro Cytotoxicity Test", the chitosan derivatives prepared in this example were tested. The results show that the cell proliferation rate of human immortalized epidermal cells is 87% under the chitosan derivative solution prepared in this example at a concentration of 2 mg / mL.

[0045] Weigh 0.7g chitosan and dissolve in 35mL dilute acid solution to obtain chitosan aqueous solution. Lysine and isoleucine (0.1:1 molar ratio) were dissolved in 15 mL of buffer solution and activated for 6 hours to obtain an amino acid activation solution with an amino acid concentration of 600 mg / mL. The amino acid activation solution was mixed with chitosan aqueous solution and reacted for 24 hours. After the reaction is finished, the product is purified to obtain the chitosan derivative with selective antibacterial properties.

[0046] According to the method requirements of GB / T 16886.5-2003 "Biological Evaluation of Medical Devices Part Five In Vitro Cytotoxicity Test", the chitosan derivatives prepared in this example were tested. The results show that the cell proliferation rate of human immortalized epidermal cells is 93% under the chitosan derivative solution prepared in this example at a concentration of 2 mg / mL.

Embodiment 3

[0051] Weigh 0.035g chitosan and dissolve in 35mL dilute acid solution to obtain chitosan aqueous solution. Arginine oligomers and leucine (0.05:1 molar ratio) were dissolved in 15 mL of buffer solution and activated for 8 hours to obtain an amino acid activation solution with an amino acid concentration of 100 mg / mL. The amino acid activation solution was mixed with chitosan aqueous solution and reacted for 24 hours. After the reaction is finished, the product is purified to obtain the chitosan derivative with selective antibacterial properties.

[0052] According to the method requirements of GB / T 16886.5-2003 "Biological Evaluation of Medical Devices Part Five In Vitro Cytotoxicity Test", the chitosan derivatives prepared in this example were tested. The results show that the cell proliferation rate of human immortalized epidermal cells is 95% under the chitosan derivative solution prepared in this example at a concentration of 2 mg / mL.

Abstract

The invention discloses a chitosan derivative with a selective antibacterial property and a preparation method of the chitosan derivative. The molecular structure formula of the chitosan derivative disclosed by the invention is of formula (1) as shown in the specification, in the formula, x, y and z are nature numbers; x, y, z are greater than or equal to 1 and less than or equal to 10000; R1 is an alkali amino acid or oligomer thereof; R2 is hydrophobic amino acid or oligomer thereof. The invention further discloses the preparation method of the chitosan derivative. The chitosan derivative disclosed by the invention is grafted with the alkali amino acid or the oligomer thereof and the hydrophobic amino acid or oligomer thereof simultaneously on a chitosan main chain; by regulating and controlling the ratio of alkali units to hydrophobic units, the chitosan derivative has certain selectivity for inhibition functions on candida albicans and lactobacillus; when the antibacterial rate ofthe chitosan derivative upon candida albicans is greater than or equal to 90%, the antibacterial rate of the chitosan derivative upon lactobacillus is less than 50%; in addition, the whole preparationprocess is energy-saving and environmental-friendly, and large-scale industrialization can be achieved.

Description

technical field [0001] The invention relates to the field of chemical modification of chitosan. More specifically, it relates to a chitosan derivative with selective antibacterial function and a preparation method thereof. Background technique [0002] In recent years, hygiene and health problems caused by antibacterial agents have frequently appeared, and people's demand for new antibacterial agents with both safety and high efficiency has become increasingly strong. As an alkaline polysaccharide, chitosan has good biocompatibility and biodegradability, and is an ideal antibacterial material. However, because the antibacterial properties of chitosan are difficult to meet the application requirements and difficult to process, derivatization is usually required to improve its antibacterial properties and water solubility. Carboxymethylation and quaternization are relatively common derivatization methods, but the chitosan derivatives prepared by the above methods are difficu...
