The invention relates to application of polygonum divaricatum, polygonum tortuosum and polygonum polystachyum total extractive, as well as elution positions thereof for preparing medicine for treatingulcerative colitis. The prepared polygonum divaricatum, polygonum tortuosum and polygonum polystachyum total extractive is ethyl alcohol aqueous extraction concentration total extractive of polygonumdivaricatum, or polygonum tortuosum or polygonum polystachyum, wherein the ethyl alcohol aqueous extraction concentration total extractive have different concentrations of 0-100%. The elution positions of the prepared polygonum divaricatum, polygonum tortuosum and polygonum polystachyum is characterized in that the ethyl alcohol aqueous extraction concentration total extractive with different concentrations of 0-100% of polygonum divaricatum, or polygonum tortuosum or polygonum polystachyum is eluted by water, 35% ethyl alcohol water, 70% ethyl alcohol water and 95% ethyl alcohol water afterbeing processed by macroporous adsorption resin to obtain a water elution position, 35% ethyl alcohol water elution position, 70% ethyl alcohol water elution position and 95% ethyl alcohol water elution position. The above elution positions of the polygonum divaricatum or polygonum tortuosum or polygonum polystachyum is used for preparing the medicine for treating ulcerative colitis.
