[0027] Systems and methods are described for analyzing biological samples mounted on transparent surfaces with multiple reagents in an automated fashion using multi-stage disposable cartridges. Multiple reagents can each be stored in separate fluid wells on the cartridge. The elastomer layers in the multi-stage disposable cartridge can be configured as fluid valves and pumps, which allow reagent fluid to flow from the wells to the sample analysis chamber where the biological sample is placed. Fluid valves and pumps can be actuated using pneumatics from the source and controlled by a computer. Thus, a biological sample can be analyzed using multiple reagents in an automated fashion. Since the fluid pathways are very small and contained within the multi-stage disposable cartridges, dead volumes are small and successive reagents can be applied in a short time. Abstract

A multi-stage disposable cartridge may have multiple fluid wells or reservoirs to hold multiple reagents and a sample viewing area to view the biological sample. Reagents can be sequentially applied to the sample by opening specific valves formed in the elastomeric flexible layer beneath the fluid wells. Fluidic channels can be formed in the first rigid plastic layer to conduct reagents to the sample. Pneumatic channels for applying suction or pressure to the valve may be formed in the second rigid plastic layer.
