Embodiment 1

[0040] This study studied the AD-related plasma metabolome based on the metabolomics method, aiming to study the pathogenesis of AD from the perspective of metabolic reprogramming and discover potential biomarkers. By collecting plasma samples from AD patients, mild cognitive impairment (MCI), non-AD neurological disease (NDC) patients and matched healthy people (Ctrl). The metabolome was analyzed by a non-targeted metabolomics method based on ultra-high performance liquid chromatography coupled with mass spectrometry. Using multivariate and univariate statistical analysis, pathway enrichment analysis to reveal metabolic changes related to AD pathogenesis and course, combined with correlation analysis to study the correlation between metabolic signatures and patients' clinical parameters.

Abstract

The present invention relates to a group of biomarkers for diagnosing AD in a subject or determining the risk of developing AD in a subject, said diagnostic markers being cholic acid, chenodeoxycholic acid, allocholic acid, indole- 3‑Lactic acid and tryptophan. The present invention also provides the application of the above diagnostic markers in the preparation of differential diagnosis reagents for Alzheimer's disease and a kit. Through the method of plasma sample collection → plasma sample pretreatment → ultra-high performance liquid chromatography mass spectrometry analysis, the plasma fingerprint analysis of AD patients, MCI patients, Ctrl and NDC populations is used to detect the contents of the above five diagnostic markers, so as to apply In the preparation of products related to the diagnosis and efficacy evaluation of Alzheimer's disease. The markers can be used for early evaluation of Alzheimer's disease, with high accuracy, fast detection speed, low cost, small trauma, and easy acceptance by patients, providing a scientific and effective treatment plan for Alzheimer's disease, which has a good application prospects.

Description

technical field [0001] The present invention relates to the technical field of biomarkers, specifically, a group of biomarkers for diagnosing AD in a subject or determining the risk of developing AD in a subject and applications thereof. Background technique [0002] Alzheimer's disease (AD) is a primary degenerative disease of the central nervous system and the most common type of dementia. The main clinical symptom of AD is slow cognitive decline, including progressive loss of memory, logical reasoning ability and language function, and finally develops into severe dementia. The biggest risk factor for AD is age. With the aging population in my country, the incidence of AD is increasing, but the pathogenesis of AD is still unclear, and there is a lack of effective clinical diagnostic markers. In addition, a large amount of evidence shows that AD has extensive pathological changes in the brain before the diagnosis of AD, that is, before the symptoms of the patient appear....
