Abstract

The invention relates to a construction system of a novel transposon mutant strain library. The construction method comprises the following construction steps that (1) a novel transposon plasmid vector in which a transposase gene tnpA is additionally added in an ITRs region is constructed; (2) a transposase gene tnpA additionally added in the ITRs region can be regulated and controlled by an arabinose promoter gene araBAD and an arabinose profilin gene araC; (3) a transposon mutant strain of different species containing a transposon is obtained; (4) the transposon mutant strain has a transposon insertion sequence at any position of a genome; and (5) the transposon mutant strain library in various forms of insert genes in a whole genome range is randomly obtained. According to the construction system, the gram-negative bacterium transposon mutant strain library covering the whole genome can be systematically and efficiently obtained, which facilitates research on genome annotation and functional analysis.
