Problems solved by technology

[0006] The present invention is to solve the above-mentioned problems, overcome the existing forecast method of observing whether the ovary is full of carapace based on experience, and cannot judge the physiological maturity and growth maturity of the ovary, so it cannot accurately predict its egg-laying time. Forecasting method By killing the parents to obtain ovarian tissue for research, it is impossible to predict the spawning period of the living parents, and provide a method for predicting the spawning period of Portunus trituberculatus

Embodiment 1

[0047] Such as Figure 1-10 shown. Tools and reagent preparation: miniature electric drill (Slitter QT-35, Ningbo Gaoqiao Slitter Tool Factory) see figure 2 , Stainless steel tweezers (GOOI-TS-14, Guangzhou Tonghui Electronic Tools Co., Ltd.) see image 3 , Jiangnan electric light source microscope (XS-212-201, Nanjing Jiangnan Yongxin Optical Co., Ltd.), C1 type visual micrometer (C1 type, Shanghai Yueci Electronic Technology Co., Ltd.), 75% alcohol (Shanghai Lingfeng Chemical Reagent Co., Ltd. ), 502 glue (Guangdong Aibida Adhesive Co., Ltd.), dry towels and cotton balls.

[0048] Live sampling: Take out the female parent of Portunus trituberculatus for breeding, dry the water on the carapace with a dry towel, and use alcohol cotton balls to disinfect the part to be drilled, that is, the front edge of the carapace and the part near the eyes. Then use a micro electric drill (a drill bit with a diameter of 1 mm, which needs to be sterilized with alcohol in advance) to dril...

Abstract

The invention relates to a method for forecasting the laying season of portunus trituberculatus. The invention samples a small quantity of ovarian tissue through drilling under the condition of not killing a parent strain and can forecast the approximate laying season of female portunus trituberculatus by the combination of visual inspection and histological observation. The key points of the method are to acquire the ovarian tissue by drilling the position which is close to the rear parts of eyes and is on the anterior border of the carapace, seal the sampling hole and adopt a modified tissue fixation liquid to fix the ovary. The method is suitable to be used by scientific research and production units and forecasts the laying season by knowing the development condition of the ovary of the parent strain in vivo. Moreover, the forecast of the laying season of the female portunus trituberculatus is favorable for the preparation of seedling facilities and baits in artificial seedling and has important significance in artificial seedling of the portunus trituberculatus.

Description

technical field [0001] The invention relates to a method for predicting the spawning period of Portunus trituberculatus, belonging to the technical field of marine culture. Background technique [0002] Portunus trituberculatus (Portunus trituberculatus) is an important marine economic crab in my country. Since the 1980s, due to overfishing and deterioration of the offshore marine environment, the natural resources of Portunus trituberculatus in my country have been declining sharply (Xue Junzeng et al., 1997 Xie Zhongming et al., 2002), therefore, since the 1990s, artificial breeding and pond culture of Portunus trituberculatus have developed rapidly in my country's coastal areas (Wu Xuqian et al., 2007). However, in actual production, due to the inability to accurately predict its spawning time has brought great difficulties to seedling production, so predicting the spawning period has very important practical significance for artificial breeding of Portunus trituberculatus...
