Embodiment 1

[0020] Since the whole genome sequence of the duck has not been tested, according to the preliminary positioning results of the QTL, through comparative genomics analysis, the homologous region on the chicken chromosome that has been sequenced is obtained, and the gene information of this region can be analyzed to find some genes as the influence Candidate genes for duck carcass traits were studied in depth, and through literature search and analysis, we selected the IGFBP7 gene that can bind IGF and insulin as the candidate gene.

[0021] According to the published chicken IGFBP7 gene sequence (NC_006091), the duck IGFBP7 gene sequence was compared and its structure was analyzed. Primers were designed according to the sequence (forward primer: 5′-ACTAAAACAGAGGTGGTCCTTC-3′; reverse primer: 5′-GAAAAGGAGCGAGTGTTAGAAC-3′), and the F1 individuals of the CAU Peking duck resource family were sequenced and analyzed to find SNPs. We found that the fourth exon and the adjacent third an...

Abstract

The invention discloses a method and a kit for testing the carcass trait of a duck flock. The method for testing the carcass trait of the duck flock comprises: detecting haptotype, which is formed by SNP, of a duck IGFBP7 gene; and judging the haptotype gene of a duck sample to be tested and determining the carcass trait of the duck. The kit provided by the invention comprises PCR amplification primers for specifically amplifying the IGFBP7 gene and other PCR amplification agents. The method and the kit of the invention have simple and quick operation, and stable and reliable result. When used in assisted breeding screening, the method and the kit have the advantages of early screening, time conservation, low cost and high accuracy. The method and the kit have a promising application prospect in meat duck breeding practice.

Description

technical field [0001] The invention relates to the technical field of molecular biology, in particular to a method and a kit for detecting carcass traits in duck populations. Background technique [0002] Carcass traits have always been important economic traits in animal husbandry production, and are also important indicators in meat duck breed selection. For a long time, meat duck carcass production has been significantly increased, mainly due to genetic selection. With the in-depth study of genetics related to poultry production traits, a large number of traits have been mapped to smaller intervals on chromosomes, laying a good foundation for the research of candidate genes and the application of subsequent genetic marker-assisted breeding. [0003] SNP (Single Nucleotide Polymorphism, single nucleotide polymorphism) mainly refers to the variation of a single nucleotide base that occurs on the genomic DNA sequence, including (1) transition (Transition), which refers to ...
