Embodiment Construction

[0017] The molecular marker-assisted breeding technology for silkworm relaxation traits provided by this embodiment comprises the following steps:

[0018] (1) Bombyx mori are reared according to conventional breeding techniques until cocooning in upper cocoons;

[0019] (2) Select the seed cocoons that need to be reserved according to the seed selection requirements, number each sample cocoon, cut a small piece of cocoon layer about 0.5 × 0.5 cm, and use the corresponding number as the sample cocoon for SDSPAGE electrophoresis of cocoon layer sericin protein;

[0020] (3) SDSPAGE sericin protein electrophoresis preparation: prepare SDSPAGE gels with acrylamide concentration of T%=5%, C%=2.6% and size of 8×8×0.1cm3, and make at least 10 plus gels on each gel Sample hole;

[0021] (4) Sample treatment: cut 10 mg of cocoon layer for each, add 300 μL of aqueous solution containing 48% (m / v) urea, 4% (v / v) mercaptoethanol, 4% (m / v) SDS, 80 ℃ water bath Treat for 5-15min. Pipett...

Abstract

The invention relates to an assisted bombyx mori breeding technology, and aims at providing a method for selecting high-reelability bombyx mori cocoons by utilizing a protein molecular marker. The method comprises the following steps: (1) preparing SDSPAGE (sodium dodecyl sulfate polyacrylamide gel electrophoresis) gel, and drilling sample holes on the gel; (2) shearing cocoon shells from breeding cocoons, adding the cocoon shells to an aqueous solution containing urea, mercaptoethanol (BME) and sodium dodecyl sulfonate (SDS), performing water bath treatment at the temperature of 80 DEG C, and absorbing samples for electrophoresis; (3) putting the electrophoresis gel and staining solution in a closed container, incubating at the temperature of 60 DEG C, washing the gel with water, adding a destaining solution, oscillating, changing the destaining solution and oscillating again; and (4) for the cocoon shells which show 110kDa protein bands in electrophoresis results, taking the corresponding breeding cocoons as sample individuals with the Sericin2 protein molecular marker. The method has the advantages that sericin protein Sericin2 is taken as the molecular marker so as to accurately and effectively select the high-reelability individuals, thus greatly improving the reelability of the bombyx mori cocoons and enhancing the breeding efficiency. For the individuals with the labeled protein Sericin2, the reelability percentage of the bombyx mori cocoons is more than 90% and the raising scale of the individuals for seed selection can be reduced by 30-50%.

Description

technical field [0001] The invention relates to silkworm assisted breeding and selection technology, in particular to a method for selecting silkworm cocoons with high unwinding properties by using protein molecular markers. technical background [0002] Raw silk and silk fabrics are the only products that account for the world's major trade share among my country's export commodities. With the profound changes in social life brought about by the continuous development of the world economy, people's requirements for clothing and other consumer goods are becoming more and more personalized, comfortable, high-quality and environmentally friendly. Silk is a natural protein fiber. Its fabric has good hygroscopicity and air permeability. Silk has very good health care effects. Clinical trials have found and proved that it is especially effective for skin itching and children's papular urticaria. The cured disease has obvious antipruritic and therapeutic effects, which makes silk...
