Problems solved by technology

The sequence and structural characteristics of the above-mentioned HIV-1 Tat molecule itself make natural Tat a candidate antigen for HIV vaccines, which still has the disadvantages of inducing immune protection, low antibody titers, and poor cross-reactivity.

Abstract

The invention relates to the technical field of biomedical engineering. Human immunodeficiency virus (HIV)-1 trans-activator of transcription (Tat) is an important regulatory protein in the early stage of HIV-1 infection. The invention aims to screen sequences of HIV-1 Tat protein mutants which are relatively stable in molecular conformation and can arouse strong immunoreactions. The amino acid sequences of the HIV-1 Tat protein mutants provided by the invention are represented by SEQ ID No.2, SEQ ID No.4 and SEQ ID No.6. The rabbit antiserums of the three Tat mutant proteins all can undergo specific reactions of the full-length Tat protein of HIV-1, the reactivity is high, and thus, the three Tat mutant proteins can be used for preparing components of anti-HIV infection polypeptide medicines or used as candidate antigens for preparing HIV Tat vaccines.

Description

Technical field [0001] The invention relates to the technical field of biomedical engineering, in particular to the human immunodeficiency virus type I Tat protein mutant sequence, and its application in preparing HIV immunogens, in preparing preventive and therapeutic HIV vaccines, and in preparing HIV testing Application in the kit. Background technique [0002] Human immunodeficiency virus (HIV) is mainly transmitted through blood, sexual contact, and mother-to-child vertical routes, and can cause acquired immunodeficiency syndrome (AIDS), or AIDS. According to new statistics from UNAIDS at the end of 2009, there are currently about 60 million people living with HIV worldwide, and 25 million people have died of AIDS-related diseases. There are currently more than 700,000 HIV-infected and AIDS patients in my country, and more than 70% of them are sexually transmitted. AIDS has become a major public health problem facing my country and the world. The current high-efficiency an...
