The method comprises the following steps of: measuring an ultraviolet absorption value of the ascorbic acid in sample extraction buffer, damaging the ascorbic acid so as to measure a background ultraviolet absorption value through an alkali treatment method, and calculating the content of the ascorbic acid through a difference. The method is characterized in that: a high-concentration alkali treatment method is adopted when a background sample is measured, so that the reduced form ascorbic acid is eliminated completely, and the influence of an alkaline environment on ultraviolet absorption of solution to be measured is avoided simultaneously. The method is simple to operate, the measurement result is accurate, and the reducing substances are not easy to get disturbed. The method is suitable for measuring sample ascorbic acid with higher numbers in botanical studies.
