[0023] 1. Isolation, Purification and Preservation of Rhizobia

[0024] Select the large and full root nodules on the roots of robust black locust from the artificially planted black locust in Guishan, Danjiangkou City, Hubei Province, carefully cut off the root nodules and some roots with scissors, and put 5-15 root nodules obtained from the root of the same black locust into a dry container. Color-changing silicone in a vial covered with absorbent cotton. Then the collected nodules were fully soaked in sterile water for imbibition, soaked in 95% alcohol for 30 s, then sterilized with 0.1% mercury chloride for 5 min, and rinsed with sterile water for 6 times. Put them into a sterile 2-ml centrifuge tube after numbering, grind the root nodules with a sterile grinding rod, add 1 ml of sterile water with a pipette and suck and drain the homogenate 3 times to suspend the homogenate, and serially dilute the suspension 10 times, 100 times and 1000 times, and then pipette 100 μl of...

Abstract

The invention relates to mesorhizobium KDRM295 which is separated from acacia root nodule, contains ACC deaminase, and can efficiently nodulate and promote growth of the acacia, and an application thereof. The mesorhizobium KDRM295 is now preserved in China Center for Type Culture Collection (CCTCC) with the preservation number of CCTCCNO: M2012331. The mesorhizobium KDRM295 contains ACC deaminase which can decompose ACC into alpha-ketobutyrate and NH3, reduce the level of plant cell for synthesizing ethylene, lighten the inhibition effect of ethylene to infection of the root nodule, improve the nodulation rate of root nodule and acacia and the level of symbiotic nitrogen fixation, provide high-level nitrogen for the growth of the acacia on the barren wasteland without fertilization, promote the growth of acacia seeding, and increase mature biomass for the acacia, thereby realizing high yield of the acacia mature with low inoculation investment, and playing a role of raising seeding and afforestation for acacia.

Description

technical field [0001] The invention belongs to the field of applied microbiology, and specifically relates to a bradyrhizobium KDRM295 isolated from root nodules of Robinia pseudoacacia, having ACC deaminase capable of forming nodules efficiently and promoting the growth of Robinia pseudoacacia, and an application thereof. The strain has been preserved in China Center for Type Culture Collection (CCTCC), the preservation number is CCTCC NO: M 2012331, and the strain name is Bradyrhizobium KDRM295. Background technique [0002] Some prokaryotic microorganisms in nature can synthesize nitrogenase, which can convert N in the air to 2 reduced to NH 3 . The nitrogen fixed by nitrogen-fixing microorganisms accounts for about 65% - 70% of the combined nitrogen on the surface. Among them, the symbiotic system of rhizobia and leguminous plants has the strongest nitrogen fixation ability, accounting for more than 65% of the biological nitrogen fixation. [0003] It is a classic ex...
