The invention provides a genetic modification method and application of a controllable
toxin of a high-targeting
metarhizium anisopliae insecticide which is toxic
in vivo and
in vitro for pests, and belongs to the technical field of
genetic engineering. An endogenous
promoter MAC05223 of a stable and high-
expression gene in
metarhizium anisopliae is screened and is used for driving efficient expression of a
toxin gene MAC08119 to construct a genetic modification strain, so that an efficient biocontrol strain with remarkably enhanced
toxicity, higher insecticidal efficiency and controllable
toxicity expression is obtained, exogenous
gene segments and exogenous toxins are not introduced, and the method is suitable for industrial production. According to the invention, high interspecific specificity and
ecological safety of
metarhizium anisopliae to locusts are maintained, metabolic burden and environmental non-target biotoxic side effects possibly brought by meaningless expression of toxins in a non-infection state are effectively avoided, host specific
pathogenicity is improved, good targeting property, safety and energy utilization efficiency are achieved, and the method is suitable for industrial production. The method can be used for improving the existing
locust biopesticide preparation, and has huge application potential in the field of green
locust prevention and control.
