Embodiment Construction

[0015] A method for directly reproducing seedlings of lotus leaves based on root buds:

[0016] 1 Materials and methods

[0017] 1.1 Source and handling of materials

[0018] The root buds of mountain lotus leaves are collected from the north slope of Changbai Mountain National Nature Reserve. After washing the soil on the root buds with tap water, rinse them with 75% ethanol (volume fraction) for 20 s on the ultra-clean workbench, soak them in 5.0% sodium hypochlorite solution (mass fraction) for 8 min, and sterile Rinse with water 10 times, then blot the surface moisture with sterile filter paper, remove the damaged tissue and use it as an explant for later use.

[0019] 1.2 Induction and rooting of clustered buds from root buds of mountain lotus leaves

[0020] Components and dosage of basic medium: 450 mg·L -1 (NH 4 ) 2 SO 4 , 1230 mg·L -1 KNO 3 , 227 mg·L -1 MgSO 4 ·7H 2 O, 346mg L -1 K H 2 PO 4 ;9.2mg L -1 FeSO 4 ·7H 2 O, 12.5mg L -1 Na 2 ·EDTA·2H 2 ...

Abstract

The invention relates to a plant propagation technology, and particularly relates to a direct umbrellaleaf direct regeneration seedling-raising method based on a radical bud. The method comprises the following steps of: 1, treating an umbrellaleaf radical bud to be used as an explant for later use; 2, inducing and rooting multiple shoots of the umbrellaleaf radical bud, to be specific, adding 0.02-0.03mg.L<-1> of naphthylacetic acid, 0.15mg.L<-1> of 6-benayl aminopurine, 0.50-0.60mg.L<-1> of kinetin, 2.20-2.40mg.L<-1> of gibberellin in a basic culture medium; and 3, rapidly propagating a umbrellaleaf sprout. The direct umbrellaleaf regeneration seedling-raising method is used for rapidly propagating umbrellaleaf by using a mode of inducing the radical bud to directly regenerate the multiple shoots and rooting, and can be applied to factorized propagation of the umbrellaleaf sprout and the large-scale cultivation and development and utilization of the umbrellaleaf. The umbrellaleaf radical bud is highest in direct regeneration rate, and the average regeneration rate reaches above 100 percent; and the acclimatization and transplant survival rate reaches above 98.0 percent.

Description

[0001] technical field [0002] The invention relates to a plant plant technology, that is, a method for directly regrowing seedlings of lotus leaves based on roots and buds. Background technique [0003] In the prior art, lotus leaf Astilbe tabularis (Hemsl.) Engler, also known as Buddha Umbrella, Big Leaf, and Big Neck Stem, is a perennial herbaceous plant of the saxifrage family. It is designated as an endangered species in the "List of Rare and Endangered Plants in China (Volume I)", and it is a national third-level key protected plant. It is designated as a provincial first-class key protected plant in the "Interim Regulations on the Protection and Management of Wild Animals and Plants of Jilin Province". Its root and whole herb are used as medicine to treat diarrhea. The leaves are large and the flowers are pure white, which can be used for greening the ground under the forest. Rhizome starch, tannin, can be used for wine making. The stems and leaves are high-quali...
