Embodiment 1

[0032] The cultivation of embodiment 1 Cordyceps militaris fruiting body

[0033] 1. Strain: The strain number obtained by independent screening is HX-64 Cordyceps militaris strain (purchased from the Biology Center of Beijing Academy of Agriculture and Forestry Sciences).

[0034] 2. Culture medium for liquid shake flasks (1L): 300g peeled potatoes (boiled juice), 10g glucose, 2g peptone, 0.5g potassium dihydrogen phosphate, 1g magnesium sulfate, 1000ml water, pH7.2.

[0035] 3. Nutrient solution for fruiting body production: 2% soybean, 1% glucose, 5% milk powder, 0.5% potassium dihydrogen phosphate, 0.05% magnesium sulfate, vitamin B110mg / 1000ml to make a nutrient solution, and adjust the pH to 5.6-7.2.

[0036] 4. The culture box made of PP material (height 10cm, upper bottom diameter 12cm, lower bottom diameter 8cm). 40g of every box of rice and 56g of fruiting body production nutrient solution are packed and mixed, sterilized and inoculated.

Embodiment 2

[0042] Example 2 Collection and preservation of ascospores of Cordyceps militaris (1)

[0043] (1) Preparation of polyspore carrier adsorption sand:

[0044] Sieve the river sand with 60 mesh, discard large particles and impurities, and then sieve with 80 mesh to remove fine sand. Use a magnet to absorb the iron, put it in a container and soak it with 10% hydrochloric acid, pour off the hydrochloric acid after 24 hours, wash it with water several times to neutral, and dry the sand.

[0045] (2) Sterilization and drying of polyspore collection containers:

[0046] Take about 25g of the treated polyspore carrier adsorption sand and mix it with an equal amount of distilled water, put it into a 250mL Erlenmeyer flask, and then sterilize the whole at 121°C for 30min with moist heat. After sterilization, put it in a drying oven at 80°C for 24 hours until completely dry.

Embodiment 3

[0056] Example 3 Collection and preservation of Cordyceps militaris ascospores (2)

[0057] (1) Preparation of polyspore carrier adsorption sand:

[0058] Sieve the river sand with 60 mesh, discard large particles and impurities, and then sieve with 80 mesh to remove fine sand. Use a magnet to absorb the iron, put it in a container and soak it with 15% hydrochloric acid, pour off the hydrochloric acid after 24 hours, wash and soak it with water several times to neutral, and dry the sand.

[0059] (2) Sterilization and drying of polyspore collection containers:

[0060] Take about 50g of the treated polyspore carrier adsorption sand and mix it with an equal amount of distilled water, put it into a 250mL Erlenmeyer flask, and then sterilize the whole at 121°C for 30min with moist heat. After sterilization, put it in a drying oven at 80°C for 48 hours until completely dry.

Abstract

The invention provides a collection and storage method of cordyceps militaris ascospores. The collection method comprises the following steps: adding sand into a multi-spore separating and collecting container which can be sealed; sterilizing and drying; putting the selected matured sporocarp of cordyceps militaris ascocarps into the multi-spore separating and collecting container containing the sand; vibrating the multi-spore separating and collecting container so as to collect the sand containing matured ascospores which are ejected out or not; sealing the collected sand in a dry and sterile container; and preserving the container at room temperature by keeping out of the sun or in a refrigerator in a cold storage manner. According to the collection method, the operation is simple and the ascospore collecting rate is increased; the preserved ascospores can retain the inheritable characters of the original parents of cordyceps militaris after being activated and novel breeds for stress resistance can be screened.

Description

technical field [0001] The invention belongs to the technical field of Cordyceps militaris cultivation, and in particular relates to a method for collecting and preserving ascospores of Cordyceps militaris. Background technique [0002] Cordyceps militaris (L. exFr) Link, also known as Cordyceps militaris and Cordyceps militaris, parasitizes on the pupal bodies of insects such as Lepidoptera, Coleoptera, and Diptera. It grows in low-altitude plains and is mainly distributed in Northeast, North China, and Northwest China. Modern research shows that Cordyceps militaris contains cordycepin (3'-deoxyadenosine), cordycepic acid (D-mannitol), adenosine, cordyceps polysaccharide, ergosterol, superoxide dismutase (SOD), selenium (Se), etc. chemical composition. It has the effects of improving human immune function, anti-cancer, anti-bacterial, anti-fatigue, anti-aging, anti-convulsions, hypoxia resistance, sedation, aphrodisiac and kidney strengthening. At the same time, it also ...
