Problems solved by technology

Although the in vitro culture system of white cotton ovules is relatively mature, the in vitro culture system of colored cotton ovules is rarely reported due to the obvious variety differences in the in vitro culture system of cotton ovules

Embodiment 1

[0021] 1. In vitro culture of brown cotton ovules

[0022] Brown cotton (Zongcaixuan No. 1) is planted in the field and managed routinely. At about 9:00 in the morning, the flowers of the day will be listed. Pull off the corolla of the young boll 1 day after flowering collected on the same day, and peel off the calyx, leaving only the flower stalk of an appropriate length. These young bells were washed twice with distilled water and put into an ultra-clean workbench, sterilized with 75% alcohol for 10 minutes, soaked in 0.1% mercuric chloride for 5 minutes, rinsed with sterile water for 6 times after disinfection, and stripped the ovules. The ovules were inoculated on BT medium with a hormone ratio of GA 3 1.0μmol / L+IAA1.0μmol / L+ZR1.0μmol / L. About 20 ovules from the same ovary were inoculated in each bottle, placed at (31±1)°C for dark culture and observed for growth.

Embodiment 2

[0035] 1. In vitro culture of green cotton ovules

[0036] Green cotton (Green Cotton No. 1) is planted in the field and managed routinely. At about 9:00 in the morning, the flowers of the day will be listed. Pull off the corolla of the young boll 2 days after flowering collected on the same day, and peel off the calyx, leaving only the flower stalk of an appropriate length. And these young bells are put into ultra-clean bench with distilled water washing 2 times. Disinfect with 75% alcohol for 10 minutes, then soak in 0.1% mercuric chloride for 10 minutes, rinse with sterile water for 7 times after disinfection, strip the ovules in the ultra-clean workbench, and inoculate the ovules in the hormone ratio of GA 3 1.0μmol / L -1 +IAA5.0μmol / L+ZR2.5μmol / L on BT medium. About 20 ovules from the same ovary were inoculated in each bottle, and about 20 bottles were inoculated for each treatment, and placed at (31±1)°C for dark culture and observed growth.

Abstract

The invention discloses a colored cotton ovule in vitro culture method, which comprises the steps of: collecting 0-2 days old young bolls of green cotton or brown cotton of the day, pulling off the corolla, stripping calyxes, and retaining flower stalks; washing the young bolls with distilled water twice, conducting disinfection with 75% alcohol for 5-15min under a sterile condition, then performing soaking with 0.1% mercury chloride for 5-15min, and conducting washing with sterile water 5-8 times, and stripping ovules; inoculating the ovules under a sterile condition into a BT medium containing hormone ZR, GA3 and IAA, inoculating 15-25 ovules of one ovary into each bottle, and carrying out dark culture for 30-40d at 30-32DEG C. Preferably, during inoculation of green cotton ovules, the hormone ratio wais GA31.0micromol / L+IAA5.0micromol / L+ZR2.5micromol / L, and during inoculation of brown cotton ovules, the hormone ratio is GA31.0micromol / L+IAA1.0micromol / L+ZR1.0micromol / L. The method provided by the invention can reduce the contamination rate, improve the survival rate, and promote cotton fiber growth and pigment synthesis.

Description

technical field [0001] The invention relates to the field of plant tissue culture, in particular to a method for in vitro culture of colored cotton ovules. Background technique [0002] Naturally colored cotton is a special type of cotton that contains natural pigments in the cotton fibers. Cotton fibers are thick-walled tubular single-cell hairs formed by the hair cells in the epidermal cells of the outer integument through processes such as protrusion, elongation, and secondary wall thickening; taking cotton fibers under field plant conditions as the research object, the results are often It is difficult to explain due to the influence of unknown factors. Kim and Triplett have pointed out that in vitro cultivation of cotton fibers allows for physiological and biochemical experiments that cannot be performed in greenhouses and fields. The effects of nutrition, low temperature, light, plant growth regulators, and other chemicals on the initiation and elongation of cotton f...
