Embodiment 1

[0023] Embodiment 1: the isolation of bacterial strain

[0024] (1) Sample collection

[0025] From June to August, healthy cotton plants were selected, and the middle and upper parts of the main stem and fruiting branches were taken 15 cm.

[0026] (2) Isolation of strains

[0027] First, the collected samples (cotton stalks) are washed with clear water, peeled off, placed in a sterilized petri dish with a diameter of 9 cm, and sterilized with 75% alcohol for 1 min. Then disinfect with 5.0% sodium hypochlorite for 60 seconds, and wash with sterilized water for 3 times, each time not less than 3 minutes. Then use sterilized absorbent paper to dry the water on the surface, cut into 0.4cm long pieces with sterilized scissors, place them in a 9cm petri dish, and incubate at 25°C. Use the last cleaning solution as a control to check whether the disinfection is thorough. Check the growth of endophytic fungi every day, and find that there are colonies growing out, and transfer t...

Embodiment 2

[0035] Embodiment 2: The inhibitory effect of CEF-082 on Verticillium dahliae of cotton

[0036] 1. Test method

[0037] The inhibitory effect of non-volatile metabolites of CEF-082 on Verticillium dahliae was determined by disc filter method. On a PDA plate with a diameter of 9 cm, spread double-layer cellophane (nitrocellulose membrane) with a diameter slightly larger than 9 cm, inoculate CEF-082 bacterial blocks (5 mm in diameter) in the center of the plate, and cultivate in a constant temperature incubator at 25°C for 7 days Remove the cellophane, inoculate Verticillium dahliae Vd080 (5mm in diameter) in the center of the plate, and cultivate in a constant temperature incubator at 25°C for 15 days. The PDA plates without CEF-082 metabolites were inoculated with Verticillium dahliae Vd080 as a control. Each treatment was repeated 4 times, and the diameter of the colony was measured by the cross method, and the inhibition rate was calculated.

Embodiment 3

[0045] Example 3: The control effect of CEF-082 on cotton verticillium wilt

[0046] 1. Test methods Two methods were used in the test to prove the control effect of CEF-082 on cotton Verticillium wilt.

[0047] pre-inoculation method. After CEF-082 was shaken and cultivated in Chapic liquid medium at 25°C and dark conditions for 7 days, it was filtered with four layers of gauze, and then filtered with a 0.22 μm microporous membrane filter to remove bacteria and spores to obtain the culture filtrate of CEF-082 spare.

[0048] Mix the sterilized vermiculite and sand at a ratio of 6:4, then make a bottomless paper bowl with a diameter of 6cm, and place it in a plastic tray of 45cm×35cm. The susceptible variety Jimian 11 was used as the test variety, and the delinted cotton seeds were soaked in clean water for 12 hours, then drenched and ready for sowing. 8 cotton seeds were sown in each pot, and 5 seedlings were retained after emergence. There were 15 pots per treatment, and ...

Abstract

The invention relates to phytopathology, in particular to cotton fungal endophyte CEF-082 and an application of the cotton fungal endophyte CEF-082 in prevention and treatment of cotton verticillium wilt. The preservation serial number of the cotton fungal endophyte CEF-082 is CGMCCNo.11313; the suppression rate of the metabolite of the CEF-082 to colony growth of the cotton verticillium wilt reaches 100%, and the suppression rate to the conidium yield is 70.73%. Greenhouse experimental results show that cotton is pre-inoculated through CEF-082 culture filtrate when true leaves of cotton seedlings appear for the first time, and the control effect on the cotton verticillium wilt is 62.37%; the corn sand grain culture of the CEF-082 is connected into a matrix at the percentage of 2%, and then the cotton seedlings are cultivated, so that the control effect on the cotton verticillium wilt is 66.88%. Compared with bacillus subtilis which is greatly used in production, the control effect is increased by 30.7% on average; the expressions of defense related genes beta-1,3-glucanase, POD, PPO, PAL and the like are improved by inoculating the cotton through the CEF-082, hence, the resistance of plants to the cotton verticillium wilt is enhanced, and the damage of pathogenic bacteria is reduced.

Description

technical field [0001] The invention relates to plant pathology, in particular to cotton endophytic fungus CEF-082 and its application in the control of cotton verticillium wilt. Background technique [0002] Cotton is an important strategic material related to the national economy and people's livelihood. Cotton Verticillium wilt is a soil-borne vascular fungal disease caused by Verticillium dahliae. It is the most important disease in cotton production in my country and even in the world. Occurrence, generally 15% -30% reduction in production, severe disease field production reduction of more than 50%, or even extinction. Due to the difficulty of crop rotation and the lack of disease-resistant varieties and pesticides with good control effects, the control of Verticillium wilt in production is almost blank, and the occurrence and harm of the disease are becoming more and more serious, becoming one of the main limiting factors for the development of cotton production.
