Embodiment 1

[0026] 1. A method for rapid detection of pyrethroid pesticides based on chicken liver esterase. The detection object is pyrethroid pesticides (fenvalerate), which is detected by a colorimetric method based on chicken liver esterase.

[0027] 1. Detection principle: Pyrethroid pesticides are hydrolyzed by chicken liver esterase to obtain the common product m-phenoxybenzaldehyde, and under acidic potassium permanganate conditions, m-phenoxybenzaldehyde is oxidized to m-phenoxybenzaldehyde Benzoic acid, the purple-red solution fades, and as the concentration of the hydrolyzate increases, the absorbance value of the solution decreases ( figure 2 , image 3 ).

[0028] 2. Detection of pyrethroid pesticides: Shred the Chinese cabbage sample, add 5ml of acetonitrile for single solvent extraction for 1min, purify the extracted solution with QuEChERS reagent pack, centrifuge and take the supernatant for later use; the volume ratio is 1:1 The ratio of :25 is to take the pesticide sa...

Embodiment 2

[0030] Embodiment 1 required chicken liver esterase preparation method, the steps include:

[0031] (1) Extraction of enzyme solution

[0032] In this example, chicken liver esterase was added with a pH-neutral buffer in a ratio of 1:3, and the tissue was broken and centrifuged to obtain a crude chicken liver esterase liquid;

[0033] (2) Partial purification of enzyme

[0034] Use 30% and 80% saturated ammonium sulfate solutions to carry out fractional precipitation of the chicken liver esterase crude enzyme solution, and then use ultrafiltration to remove small molecular salts;

Abstract

The present invention provides a method for rapid detection of pyrethroid pesticide residue. The method comprises the following steps: hydrolyzing a pyrethroid pesticide with known concentration; detecting coloration: determining the absorbance values at 525nm of a control pyrethroid pesticide and the pyrethroid pesticide with known concentration by a spectrophotometry method to obtain a linear relationship standard curve between the absorbance value difference and the concentration of pyrethroid pesticide; hydrolyzing a pyrethroid pesticide with unknown concentration, detecting the sample by using the coloration detection system method to obtain the absorbance difference of the sample based on carboxylesterase hydrolysis coloration method; and determining the content of pyrethroid pesticide in the sample by using a standard curve method. The method of the invention can be widely used in the qualitative and quantitative detection of type I and type II pyrethroid pesticide residues in food, agricultural products such as vegetables and fruits, and environment, has the advantages of simple and fast procedure and low cost, and is easy to be used in rapid detection equipment in the field.

Description

technical field [0001] The invention belongs to the field of agriculture, and relates to a pyrethroid pesticide, in particular to a method for rapidly detecting pyrethroid pesticide residues. Background technique [0002] Pyrethroid pesticides are a class of artificially synthesized insecticides. Their broad-spectrum insecticidal effects and low toxicity to mammals have gradually replaced traditional organophosphorus pesticides. Recent studies have found that a certain residual dose can cause serious damage to the central nervous system of exposed organisms. Therefore, research on new detection methods for pyrethroid pesticide residues has received extensive attention. At present, the commonly used analytical method for pyrethroid pesticide residues is instrument detection, which requires expensive analytical instruments and requires high professional skills for analysts; the enzyme inhibition method is only sensitive to organophosphorus pesticide residues and is not suit...
