Embodiment 1

[0022] Example 1 Up-regulated expression of MIR3945HG V1 in MΦs infected with MTB

[0023] Isolation of PBMC from healthy volunteers, immunomagnetic separation of CD14 + For monocytes, GM-CSF was used to induce monocytes to differentiate into MΦ, and the obtained MΦ was infected with MTB. After 72h, real-time fluorescent quantitative PCR was used to detect the expression level of MΦ in MΦ.

[0024] Test results such as figure 1 As shown, it can be seen that the expression level of MΦ of MΦ after MTB infection is significantly up-regulated, indicating that MTB infection promotes the expression of MIR3945HGV1.

Embodiment 2

[0025] Example 2 Detection of expression of MIR3945HG V1 in PBMCs of patients with bacterial negative pulmonary tuberculosis by real-time fluorescent quantitative PCR

[0026] The RNA of PBMC of 30 patients with bacterium-negative pulmonary tuberculosis and 32 healthy volunteers was extracted, and the expression level of lncRNA-MIR3945HGV1 was detected by real-time fluorescent quantitative PCR.

[0027] Test results such as figure 2 As shown, it can be seen that the expression of MIR3945HG V1 in PBMCs of patients with bacterial-negative pulmonary tuberculosis showed a significant up-regulation trend (P<0.0001).

Embodiment 3

[0028] Example 3 MIR3945HGV1 as a new target for the diagnosis of bacterial negative pulmonary tuberculosis

[0029] This study found that, compared with healthy people, the expression of MIR3945HG V1 in PBMCs of tuberculosis patients showed a very significant up-regulation trend (P<0.0001). In order to understand whether the expression change of MIR3945HGV1 in PBMC can be used to distinguish healthy people from tuberculosis patients, we used ROC curve to analyze whether the expression change of MIR3945HGV1 in PBMC can be used for the diagnosis of tuberculosis.

[0030] According to the relative expression of MIR3945HG V1 in the PBMC of healthy people and tuberculosis patients, statistical analysis software was used to establish the ROC curve with specificity on the abscissa and sensitivity on the ordinate, see image 3 . The results showed that the area under the ROC curve AUC(A)=0.9258, indicating that MIR3945HGV1 has a high accuracy in the diagnosis of tuberculosis patient...

Abstract

The invention discloses an application of lncRNA (long noncoding RNA)-MIR3945HG V1 in diagnosis of smear and culture negative pulmonary tuberculosis. The invention finds that up-regulated expression of MIR3945HG V1 is caused by MTB (mycobacterium tuberculosis) infection with Mphi; results of PBMC detection of 30 patients with smear and culture negative pulmonary tuberculosis show that the expression levels of MIR3945HG V1 of the patients are obviously higher than those of healthy volunteers; an analysis result of a receiver operating characteristic curve shows that MIR3945HG V1 is expected to become a novel target for diagnosis of smear and culture negative pulmonary tuberculosis. Uninfected people and the patients with smear and culture negative pulmonary tuberculosis can be distinguished effectively by detecting the expression level of MIR3945HG V1.

Description

technical field [0001] The present invention specifically relates to the application of lncRNA-MIR3945HGV1 in the diagnosis of bacterial negative pulmonary tuberculosis. Background technique [0002] Tuberculosis is still the infectious disease with the highest infection rate in the world, and the prevention and control situation is very severe. Rapid and sensitive diagnosis is the key to improving the treatment effect and effectively controlling the spread of tuberculosis. Among the pulmonary tuberculosis patients in China, bacterium-negative pulmonary tuberculosis patients account for 60% to 70% of active pulmonary tuberculosis patients, and the clinical symptoms and imaging features of many other lung diseases are difficult to distinguish from tuberculosis. There is a lack of effective diagnostic methods for bacterial-negative pulmonary tuberculosis, which often results in delayed or misdiagnosed patients. Although my country has formulated some diagnostic criteria, the...
