[0026] refer to Figure 1 to Figure 6 , a method for preparing a bacterial biosensor for antibiotic detection and analysis in the environment, comprising the following steps:

[0027] (1) Surface functionalization of alumina nanoporous membrane:

[0028] After the aluminum oxide nanoporous membrane is silanized with 3-(2,3-glycidoxy)propyltrimethoxysilane, it is sealed in a polydimethylsiloxane (PDMS) container with epoxy resin; Add the Salmonella antibody solution and let stand at 0-4°C for 3-6 hours to fix the antibody on the porous membrane;

[0029] Slowly pass in and out PBS buffer at a constant rate to the upper and lower chambers of the PDMS container to wash the unfixed Salmonella antibody for 1 to 3 times; put the prepared alumina nanoporous membrane device at 4°C for standby ;

[0030] (2) Capturing bacteria: Disperse the cultivated Salmonella in PBS buffer solution, make Salmonella solutions of different concentrations, take the above prepared Salmonella solution...

[0037] Example 1: The preparation process of bacterial biosensor is as follows figure 1 Shown: 2% 3-(2,3-epoxypropoxy) propyltrimethoxysilane silanized aluminum oxide nanoporous membrane, sealed the silanized alumina nanoporous membrane with epoxy resin glue in a polydimethylsiloxane (PDMS) container. Salmonella antibody solution (0.5 μg / mL, 100 μL) was added, and after standing at 4° C. for 4 hours, the antibody was immobilized on the porous membrane. The PBS buffer solution was slowly passed into and withdrawn at a constant rate from the upper and lower chambers of the PDMS container to wash unfixed Salmonella antibodies, and this step was repeated 3 times. The prepared alumina nanoporous membrane device was placed at 4°C for standby.

[0038] Disperse the cultured Salmonella in PBS to make Salmonella solutions with different concentrations. Take the Salmonella solution (100 μL) prepared above, add the aluminum oxide nanoporous membrane device, and after standing for 8 ho...

Abstract

The invention provides a preparation method of a bacterial biosensor for antibiotic detection analysis in an environment. An alumina nano-porous film is used as a core sensing platform, 3-(2,3-epoxypropoxy)propyltrimethoxysilane (GPMS) is used for treatment, and a bacterial antibody is directly fixed to a membrane and used for capturing corresponding salmonella. Bacteria result in blockage of a nano-porous membrane, antibiotics act on the bacteria and are used for sterilization, the blockage effect of the nano-porous membrane is reduced, and an electrochemical impedance method can be utilized for analysis detection. The invention provides the preparation method of the bacterial biosensor for rapid and simple antibiotic detection analysis in the environment.

Description

technical field [0001] The invention relates to a preparation technology of a sensor device, in particular to a preparation method of a bacterial biosensor used for the detection and analysis of antibiotics in the environment. Background technique [0002] Antibiotics are effectively used to treat a variety of bacterial infections. my country is a big country in the production and use of antibiotics, mainly in the livestock and poultry breeding industry, where antibiotics are widely used as feed additives. However, most antibiotics are not absorbed by the animal body and are excreted with urine or feces, and enter the water environment, which can lead to the production of a large number of drug-resistant pathogenic bacteria and become a new source of pollution in the natural water environment. [0003] Traditional antibiotic detection methods include gas chromatography, chromatography / mass spectrometry, high performance liquid chromatography, and capillary electrophoresis a...
