[0025] The following examples are only used to further illustrate the content of the present invention, and are not intended to limit the present invention in any respect. Those skilled in the art will appreciate that modifications may be made to the detailed description described below without departing from the scope, spirit and spirit of the invention as claimed in the appended claims.

[0026] In all the following examples, unless otherwise stated, methods, instruments, reagents and experimental schemes commonly used by those skilled in the art are used. The basal medium for differentiation was RPMI 1640 (Gibco), and the differentiation supplement was S12 (patent pending) developed by our laboratory or commercial supplement B27 (Gibco). In all the following examples, unless otherwise stated, S12 is used as the differentiation supplement.

Abstract

The invention provides a method of using multipotent stem cells to prepare epicardial cells, comprising: 1) inducing multipotent stem cells to differentiate into mesodermal cells by activating a WNT signal pathway; 2) converting the mesodermal cells in step 1) into myocardial precursor cells by inhibiting the WNT signal pathway; 3) converting the myocardial precursor cells in step 2) into preliminary epicardial cells by activating a fibroblast growth factor signal pathway and the WNT signal pathway; preparing pericardial cells with the preliminary epicardial cells of step 3). The invention also relates to: a method of using multipotent stem cells to prepare vascular smooth muscle cells and cardiac fibroblasts; epicardial cells, vascular smooth muscle cells and cardiac fibroblasts all of which are prepared through the above method; and their application.

Description

technical field [0001] The present application relates to a method for preparing epicardial cells from pluripotent stem cells, and also relates to a method for preparing vascular smooth muscle cells and cardiac fibroblasts from pluripotent stem cells, and epicardial cells, vascular smooth muscle cells and cardiac fibroblasts prepared by the above method Fibroblasts and their uses such as epicardial cells for cardiac injury repair and screening of new therapies for cardiac injury, and redifferentiation into vascular smooth muscle cells for disease and drug screening models, vascular tissue engineering. Background technique [0002] Epicardial lineage cells are required during embryonic heart development. Pre-Epicardial Cell Precursors: Pro-epicardial cells, derived from visceral mesoderm, develop in the inflow tract of the heart tube immediately posterior to the atrium and sinus venosus. The precise source of pre-epicardial cells is not well understood, but genetic lineage t...
